Gene expression profile of chicken lung in response to Avian influenza virus challenge in broiler chickens

Project description:To determine the host response to AIV in chicken lungs, a whole chicken genome array was used to analyze RNA isolated from chicken lungs infected with AIV (4dpi) or medium control (NS). Dual-color, direct comparisons were carried between AIV infected and non-infected controls. Each comparison includes four biological replicates. There were 508 mRNAs (347 down-regulated) were differentially expressed following AIV infection. From the results, MX1, IL-8, IRF-7, TNFRS19 are identified as strong candidate genes involved in regulating the host response to AIV infection in the lungs of broiler chickens. Further gene specific knock-down assay is warranted to elucidate underlying mechanism of AIV infection regulation in the chicken.

Axiom Genome-Wide Chicken Array of Broiler Chickens

Project description:DNA was isolated from whole red blood cells from various lines and crosses of broiler chickens. DNA was genotyped using Axiom genome-wide chicken array and cel files were analyzed using Axiom Analysis Suite Software (version 3.0.1) with Gallus gallus 5.0 using the software's Best Practices for agricultural animals. The results were exported (Genotyping_Data-3-21-2018.vcf) for all genotype calls and text file of all SNPs with >= 97% call rate rate was also produced for filtering the VCF file (ALL_SNPSs_with_Call_Rate_97_Plus_3-21-2018).

Dietary supplement vanadyl sulfate selectively eliminates senescent cells

Project description:During human evolution, lifestyles including dietary choices have had a major impact on shaping of human physiology and health, including aging. However, the underlying mechanisms remain largely unknown. Here, we use a blood nutrient compound library-based screening approach to demonstrate that dietary supplement vanadyl sulfate (VS) selectively induces cell death in senescent cells and ameliorates phenotypes of aging and age-related disorders in vivo. Integrated chemical biology and multi-omics studies reveal that senescent cells are specifically sensitive to VS due to reduced cysteine reactivity with accumulated glutathione disulfide. VS selectively inactivates reactive cysteines on proteins to induce formation of incorrect disulfide bonds in senescent cells with imbalanced cellular redox status, causing aberrant protein misfolding and aggregation and eventual cell death. Our findings uncover a new senolytic mechanism to mitigate age-related disorders and reshape human physiology.

Effects of chronic heat exposure on gene expression in abdominal fat of growing broiler chickens.

Project description:Exposure to chronic heat stress not only reduces body weight gain in growing broilers, but also enhances fat deposition in the abdomen, subcutaneous, and intramuscular. To elucidate this phenomenon, numerous studies have examined changes in the transcripts, protein, and metabolic levels in the liver, the primary site of de novo fat synthesis in birds. On the other hand, changes in adipose tissue, the direct site of fat deposition, have not been unclear. To elucidate the changes in adipose tissue of chickens heat-exposure, we demonstrated the experiments rearing chickens in a heat environment (32°C) for 14 days, and then compared with the control group that reared in thermoneutral condition (22°C), and a pair-fed groups that reared in thermoneutral condition but feed intake was limited to the same levels as that in the heat-exposed group. Despite the feed intake was equal to that of the pair-fed group, the ratio of abdominal fat to body weight in the heat-exposed group was maintained at the same level as that in the control group. In addition, the heat-exposed group showed a significant decrease in the concentration of free fatty acids in plasma compared with the other 2 groups. Transcriptome analysis of abdominal fat between the heat-exposed and control groups revealed 459 differentially expressed genes were indicated. Notably, in heat-exposed group, the expression of genes related to lipid synthesis increased in adipose tissue compared with control group. Taken together, these results indicate that chronic heat has a lipid-deposition-promoting effect on growing broiler chicken adipose tissue
