Project description:Soybean plants were subjected to a multifactorial stress combination of up to five different stresses (water deficit, salinity, low phosphate, acidity, and cadmium), in an increasing level of complexity. All stresses were applied at the beginning of the experiment except for water deficit stress that was imposed after 21 days. Leaves and flowers were collected from 5-7 different plants under the mentioned stress conditions and after 10 days of starting water deficit conditions. Differential gene expression compared to control was studied using RNAseq method for all the possible stress combinations.

Multiomics-based insights into tomato adaptation to multifactorial stress combination

Project description:Multifactorial stress combination (MFSC) represents a major threat to crop productivity under climate change scenarios. While the physiological effects of MFSC have been studied, its impact on gene expression and primary metabolism remains poorly understood in tomato plants. In this study, we performed an integrative transcriptomic and metabolomic analysis of tomato (Solanum lycopersicum) plants exposed to combinations of up to six low-intensity abiotic stresses. RNA-seq analysis revealed a stress complexity-dependent increase in differentially expressed genes (DEGs), including 258 core transcripts and 155 transcription factors uniquely regulated under 4-, 5-, and 6-stress conditions. Heat-specific analysis identified 103 uniquely regulated transcripts, including two key transcription factors (Zinc Finger TF32 and a B3-domain family protein) suggested as central regulators of different combinations involving heat stress. Our findings define a conserved, stress-complexity-dependent molecular architecture in tomato and identify candidate markers for improving crop resilience under MFSC.

Project description:Climate change and industrial pollution are responsible for major losses in crop yield worldwide. Tomato plants are subjected to multiple stress conditions in combination when growing in the natural field. In this work, we analyzed the transcriptomic response of tomato plants to the combination of salinity and heat stress, when plants were or were not inoculated with the beneficial bacteria Pseudomonas Putida. The results suggest that specific transcripts are regulated only under stress combination in plants inoculated with the bacteria. These transcripts could be related with the enhanced tolerance of inoculated plants under conditions of stress combination.

Project description:Plants in nature are exposed to combinations of abiotic and biotic stresses. Climate change driven increase in frequency of abiotic stress combinations is being detrimental to the plant survival and growth. Previously we found that Arabidopsis plants when exposed to combination of abiotic stresses termed as multifactorial stress combination (MFSC), the accumulation of multiple mild abiotic stresses becomes damaging to plants causing severe reduction in plant survival which otherwise not detrimental when exposed as single stresses. Here in this study, we investigated the role of a transcription factors Basic helix loop helix (bHLH35) in MFSC tolerance in Arabidopsis seedlings.

RAP2.3 is required for MYB51 and SIGMA3 expression during the response of Arabidopsis thaliana to multifactorial stress combination

Project description:In nature, plants are subjected to multiple environmental stress factors simultaneously or sequentially. Recent studies revealed that when three or more stress factors impact a plant simultaneously (termed ‘multifactorial stress combination’; MFSC), plant survival declines, even if the intensity of each individual stress involved in the MFSC is low. We previously identified RAP2.3 as a key transcription factor (TF) required for Arabidopsis thaliana survival, specifically under a MFSC of salt+excess light+heat stress (i.e., S+EL+HS). Here we report that RAP2.3 is required for the expression of SIGMA3, a nuclear-encoded factor that directs plastid RNA polymerase to specific plastid promoters, and MYB51, a key stress response TF involved in glucosinolate metabolism and oxidative stress responses, specifically during a MFSC of S +EL+HS. Like rap2.3 mutants, myb51 and sig3 mutants display significantly low survival rate specifically under the MFSC of S+EL+HS. Based on MYB51 gene regulatory network analysis and jasmonic acid (JA) mutant characterization, we further reveal that suppression of JA signaling could play an important role in promoting plant survival under conditions of S+EL+HS. Our findings uncover an additional layer of the response of plants to MFSC, as well as identify potential targets for breeding of crops with enhanced tolerance to climate change.

Microarray analysis of transcriptional responses to abscisic acid and salt stress in Arabidopsis thaliana

Project description:To investigate differences in plant responses to salt and ABA stimulus, differences in gene expression in Arabidopsis in response to salt and ABA were compared using an Agilent oligo microarray. Four-week-old Arabidopsis thaliana ecotype Columbia (Col-0) seedlings were treated with either 150 mM NaCl or 10 μM ABA for 6 hours; unstressed seedlings (control sample) were collected in parallel to avoid the possible effects of circadian rhythms. The results revealed that 31 genes were up regulated by both NaCl and ABA stress, and 23 genes were down-regulated by these stressors. To provide further validation of our microarray experiment data, ten genes from this signature were quantified in the same RNA samples by quantitative real-time PCR.

Hydrogen peroxide stress and Zat12 over-expression in Arabidopsis.

Project description:In this experiment we tested the transcriptome of transgenic Arabidopsis seedlings (5-day-old) constitutively expressing the zinc-finger protein Zat12 (At5g59820) under the control of the 35S-CaMV promoter (Zat12). The transcriptome of these seedlings was compared to that of wild type seedlings grown under the same conditions (WT) and to that of wild type seedlings grown under the same conditions and subjected to a hydrogen peroxide stress (WT+H2O2). Hydrogen peroxide treatment was performed by applying 20 mM hydrogen peroxide for 1 hour. In parallel to these experiments transgenic plants expressing Zat12 were subjected to a similar hydrogen peroxide stress (Zat12+H2O2). All treatments were performed with similar size and age seedlings grown in liquid culture (MS) and sampled at the same time as described by Davletova et al., 2005.
