Diabetic lung

Project description:Effect of type 1 diabetes (induced by streptozotocin 60 mg/kg) on lung gene expression. Wistar rats, male. At age 8 weeks control rats got IP buffer, diabetic rats got streptozotocin. At age 12 weeks animals were anesthetized and lungs removed. RNA was extracted with Trizol, and gene expression array analysis was performed using Affymetrix RAE 230A microarrays according to the directions from the manufacturer. Arrays were scanned using a Hewlett Packard Gene Array scanner, and analyzed with Affymetrix MAS 5.0 software. Expression levels reported are the output from the MAS software.

Plasma cell dyscrasias expression profiles associated with distinct IGH translocations in multiple myeloma

Project description:This series of microarray experiments contains the gene expression profiles of purified plasma cells (PCs) obtained from 7 monoclonal gammopathy of undetermined significance (MGUS), 39 newly diagnosed multiple myeloma (MM) and 6 plasma-cell leukaemia (PCL) patients. PCs were purified from bone marrow Seriess, after red blood cell lysis with 0.86% ammonium chloride, using CD138 immunomagnetic microbeads. The purity of the positively selected PCs was assessed by morphology and flow cytometry and was > 90% in all cases. 5 micrograms of total RNA was processed and hybridized to the Affymetrix HG-U133A chip following the manufacturer's instructions. After scanning, the images were processed using Affymetrix MicroArray Suite (MAS) 5.0 software to generate gene expression intensity values. Arrays normalization was performed using MAS 5.0 "global scaling" procedure, which normalizes the signals of different experiments to the same target intensity (TGT=100). Keywords: other

Project description:Natural variation in gene expression among healthy human individuals has been largely unexplored. In order to understand the genetic basis of variation in gene expression between normal human individuals, we need to obtain genome-wide expression data from various populations. Studies in monozygotic twins could enable us to estimate the size of the contribution of genetic and environmental factors to the natural variation in gene expression. In this work, we report the gene expression analysis of 5 pairs of monozygotic twins and 3 unrelated individuals Normal healthy twin pairs were recruited for the study. Three more normal individuals including two females and one male were recruited. First, labeled products were hybridized with the Affymetrix GeneChip Test3 arrays. If the results were judged satisfactory, hybridization was subsequently carried out with the HG U95A arrays as per manufacturer's instructions. Arrays were hybridized at 45°C for 16 hours. The array was then washed using an automated Gene Chip Fluidics Station 400. After washing, the array was stained with streptavidin-phycoerythrin and scanned with a HP Gene Array Scanner. The data was analyzed using Gene Chip Software for expression analysis (MAS 5). All Gene Chip experiments were performed at the Weizmann Institute of Science, Rehovot, Israel. Following comparisons were carried out: 1. Between female identical twin pair (GSM14480 and GSM14481; GSM29053 and GSM 29054; GSM 29055 and GSM 29056) 2. Between male identical twin pair (GSM14478 and GSM14479; GSM 29057 and GSM 29058) 3. Between unrelated female individuals (include GSM14483, GSM20645 also but exclude comparisons within same twin pairs) 4. Genetic variation in radiation-induced cell death

Project description:We used microarrays to measure the expression levels of genes in irradiated immortalized B cells, lymphoblastoid cells, from members of Centre d'Etude du Polymorphisme Humain (CEPH) Utah pedigrees. Data were collected for cells at baseline and 2 hours and 6 hours after exposure to 10 Gy of ionizing radiation (IR). We measured the expression levels of genes in irradiated immortalized B cells, lymphoblastoid cells, from members of 30 Centre d'Etude du Polymorphisme Humain (CEPH) Utah pedigrees (CEPH 1331, 1332, 1333, 1341, 1344, 1346, 1347, 1349, 1354, 1356, 1357, 1358, 1362, 1408, 1413, 1416, 1418, 1420, 1421, 1423, 1424, 1444, 1447, 1451, 1454, 1456, 1458, 1463, 1477, 1582). Cells were irradiated at 10 Gy in a 137Cs irradiator. Cells were harvested prior to radiation and at 2 and 6 hours following exposure to IR.
