Project description:Partial tumor irradiation combining low dose radiation with high dose radiation within the same tumor mass would prompt an antitumor effect when combined with immunomodulators. Partial irradiations (50%/50%) of subcutaneous MC38 tumors with millimetric precision to deliver low dose radiation (2Gy) and high dose radiation (16Gy) within the same tumor was performed and single-cell RNA sequencing of CD45+ cells analysis was done in both the two part of the tumor to identify tumor-infiltrating immune cells.

Melanoma B16 transcriptome sequencing in C57BL16 mice under dacarbazine treatment

Project description:On day 7 after B16 melanoma cells transplantation, C57Bl6 mice with subcutaneous palpable tumors were randomly divided into 2 groups for experimental treatment with Dacarbazine. Group 1 «Control (PBS) », n=5, animals in this group were intraperitoneally injected with phosphate buffered saline in a volume of 250 µl. Group 2 «DTIC», n=5, animals in this group were intraperitoneally injected with a solution of Dacarbazine (DTIC) (Sigma-Aldrich, USA) in phosphate buffer at a concentration of 50 mg/kg of animal weight. Injections with PBS or DTIC were carried out three times on days 8, 10 and 12 after melanoma cell transplantation, on day 14, mice were sacrificed for further analysis of primary tumors and organs.

In contrast, Ang2 overexpression promoted the EAE-induced activation of immune cells and endothelial cells. All the samples were analyzed using the Chromium Single-cell 3'RNA-sequencing system.

RNA/WES/ATAC/scRNA sequencing of murine melanoma primary and LN metastases

Project description:The purpose of this study is to identify transcriptional, mutational, andepigenetic differences between LN metastases from primary tumors. Using in vivo serial passaging of the minimally metastatic syngeneic murine melanoma cell line B16-F0, we have generated nearly 300 unique cell lines spanning nine generations of in vivo passaging, derived from LNs, and exhibiting varying degrees of metastatic potential to LNs. Here, we perform mRNAseq on 30 of the lines representing a range of anatomical locations (distinct LNs), generations, and phylogenetic lineages. The subsequent differential expression analysis reveals a marked upregulation of immune-related genes. Additionally, we perform WES to identify differences in particular mutations, total mutational burden, and predicted neoantigen burden. Additionally, we perform ATAC-seq to assess changes in chromatin accessibility hat correlate with LN metastasis. We also perform scRNA-seq on dissociated LNs from mice with and without LN metastases.

Gene expression profiles of melanoma tumor samples from patients treated with checkpoint inhibitors (mouse)

Project description:Checkpoint inhibitors have improved outcomes for patients with metastatic melanoma, but primary or acquired resistance often occurs. We used the mouse melanoma cell line B16, originally derived from C57BL6 mice, to model changes in tumor cell gene expression that occur with acquired resistance to checkpoint inhibitors. B16 cells used were engineered to express the fluorescent marker tdTomato, to assist monitoring and cell sorting. Previously-untreated B16 cells (F0) were inoculated into C57BL6 mice, followed by treatment with triple-checkpoint inhibitor therapy (3I) with combined antibodies to CTLA4, PD-1, and PD-L1. Tumors that emerged despite immune selection pressure were harvested, grown again in vitro as a cell line, and then re-inoculated into a new generation of mice. This was repeated for 4 generations (F4) until inoculations were 100% successful at generating immune-resistant tumors.

Project description:B16-BL6 mouse melanoma that had been maintained in C57BL/6J mice were used to evaluate the 5-aminolevulinic acid (ALA) and radiation dose effects on radiotherapy. Mice were divided into 6 groups after implantation of B16-BL6 cells; (1) no treatment (NT) ; (2) 5-ALA treatment (ALAT); (3) 10 session of fractionated irradiation (2Gy/day) (20XT); (4) 10 sessions of 5-ALA treatment followed by fractionated irradiation (2Gy/day) (ALA-20XT); (5) 10 session of fractionated irradiation (3Gy/day) (30XT); (6) 10 sessions of 5-ALA treatment followed by fractionated irradiation (3Gy/day) (ALA-30XT).

Radiation-induced epigenetic DNA methylation modification of radiation-response pathways

Project description:Treatment of tumors with ionizing radiation for cancer therapy induces biological responses that include changes in cell cycle, activation of DNA repair mechanisms, and induction of apoptosis or senescence programs. What is not known is whether ionizing radiation induces an epigenetic DNA methylation response or whether epigenetic changes occur in genes in pathways classically associated with the radiation response. We exposed breast cancer cells to 0, 2, or 6 Gy and determined global DNA methylation at 1, 2, 4, 8, 24, 48, and 72 hours post-irradiation. We found that radiation treatment resulted in a DNA methylation response and that cell cycle, DNA repair, and apoptosis pathways were enriched in genes are were differentially-methylated. DNA methylation profiling of ionizing radiation treated cells using the Infinium HumanMethylation450 BeadChip.
