Project description:Disease resistance is associated with a plant defense response that involves an integrated set of signal transduction pathways. Gene Expression studies were performed using Arabidopsis wild type seedlings treated with and without salicylic acid (SA). In this study we examined pathogenesis related genes are upregulated. We used Affymetrix GeneChip to study the genome wide gene expression with and without induction of defense by treatment with SA Experiment Overall Design: Arabidopsis seedlings Col-0 were grown for 9 days in controlled environment rooms at 22°C temperature and a photoperiod of 16 h light 100 µm m2 s_1 cool-white fluorescent illumination. After 9 days seedlings were transferred into water as a mock treatment and with the defense related signaling molecules salicylic acid (SA) for one day. The concentration of SA (Sigma, St. Louis) was 2mM and pH 6.5. RNA extracted from SA and mock treated seedling and hybridize with Affymetrix GeneChip to examine the change in plant defence gene expression due to induction of SA.

Project description:Reactive oxygen species such as hydrogen peroxide (H2O2) are important in biotic and abiotic stress responses in plants, but their source is often unclear. We have identified an Arabidopsis mutant that shows loss of stress responsive GSTF8 gene expression in response to the plant defence signal salicylic acid (SA) . The mutant showed increased susceptibility to both fungal and bacterial pathogens. The dsr1 mutation was mapped to mitochondrial succinate dehydrogenase (SDH1-1) and dsr1 had reduced SDH activity and a lowered mitochondrial H2O2 production. To better understand the loss of SA-response in dsr1, we performed ATH1 microarray analysis in WT and dsr1 10 hours after a 1 mM SA treatment, a point at which we observed maximal SA-induced GSTF8 promoter activity. 4 day old WT (a Columbia line containing a -800GSTF8::LUC reporter) and dsr1 seedlings (a mutant Columbia line containing a -800GSTF8::LUC) reporterwere treated with water (mock) or 1 mM SA for 40 min. Ten hours after treatment, samples were collected.

Project description:Subgroup Ib BHLH genes are induced by Fe deficiency. BHLH038 and BHLH039 were shown to be connected with salicylic acid. The triple knockout 3xbhlh (bhlh039-1 bhlh100-1 bhlh101-1) shows a stronger leaf chlorosis at - Fe than the wild type. The responses of this triple mutant were studied at - Fe in comparison to the wild type in the presence and absence of salicylic acid (SA). Wild type and 3xbhlh seedlings (bhlh039-1 bhlh100-1 bhlh101-1) were grown at M-bM-^@M-^S Fe and treated for six hours with or without 100 M-BM-5M SA. Three biological replicates were generated.

Project description:Salicylic acid (SA) is a plant defense hormone required for immunity. Arabidopsis NPR1 and NPR3/NPR4 were previously shown to bind SA and proposed as SA receptors. However, unlike NPR1, loss of NPR3/NPR4 does not block SA-induced defense gene expression. Here we report that NPR3/NPR4 function as transcriptional repressors and SA inhibits their activities to promote the expression of key immune regulators. npr4-4D, a newly identified gain-of-function allele that renders NPR4 unable to bind SA, constitutively represses SA-induced immune responses. In contrast, the equivalent mutation in NPR1 abolishes its function in promoting SA-induced defense gene expression. Further analysis revealed that npr4-4D and npr1-1 have additive effect on blocking SA-induced defense gene expression, suggesting that NPR4 and NPR1 function in parallel to regulate SA-induced immune responses. Our study reveals the molecular functions of SA receptors NPR3/NPR4 and uncovers a brand new mechanism of SA perception.

Project description:Infection by the pathogen grape powdery mildew (Erysiphe necator) causes changes in the transcriptome of its susceptible host Vitis vinifera. Infection triggers the host to synthesize the signaling molecule salicylic acid (SA) which regulates the expression of a broad range of defense-related plant genes. In addition, it is hypothesized that E. necator directly modulates gene expression in V. vinifera via the haustorial complex. This microarray experiment was designed to dissect host transcriptome changes triggered directly by E. necator infection and indirectly through the SA response. We accomplished this by conducting two separate global leaf transcriptome analyses using the Vitis Affymetrix GeneChip platform: in one, we compared the leaves with fully established PM colonies to healthy reference leaves, in another, we compared healthy leaves with artificially elevated SA levels to healthy reference leaves. Overlaying host transcriptome changes from these two experiments enabled us to glean out V. vinifera genes that modulate their expression in response E. necator in an SA-independent manner.

Project description:Nonsteroidal anti-inflammatory drugs (NSAIDs), including salicylic acid (SA), target mammalian cyclooxygenases. In plants, SA is a defense hormone that regulates NON-EXPRESSOR OF PATHOGENESIS RELATED GENES 1 (NPR1), the master transcriptional regulator of immunity-related genes. We identify that the oxicam-type NSAIDs tenoxicam (TNX), meloxicam, and piroxicam, but not other types of NSAIDs, exhibit an inhibitory effect on immunity to bacteria and SA-dependent plant immune response. TNX treatment decreases NPR1 levels, independently from the proposed SA receptors NPR3 and NPR4. Instead, TNX induces oxidation of cytosolic redox status, which is also affected by SA and regulates NPR1 homeostasis. A cysteine labeling assay reveals that cysteine residues in NPR1 can be oxidized in vitro, leading to disulfide-bridged oligomerization of NPR1, but not in vivo regardless of SA or TNX treatment. Therefore, this study indicates that oxicam inhibits NPR1-mediated SA signaling without affecting the redox status of NPR1.

Comparison of transcriptomes between hybrids exhibiting heterosis for disease resistance and their parents after inoculation with Pst DC3000 revealed that several key genes involved in salicylic acid (SA) biosynthesis were significantly up-regulated in hybrids. Consistently, in response to bacterial infection, more SA [both free SA and SA glycoside (SAG)] accumulated in hybrids compared with both parents. In addition, heterosis for bacterial defense was significantly compromised in hybrids of pad4 mutants in which the SA biosynthesis pathway was blocked. Moreover, we further revealed that increased histone H3 acetylation of the key genes involved in the SA biosynthesis pathway correlated with their up-regulated expression in hybrids.
