Ecotoxicogenomic analysis on the soil nematode Caenorhabditis elegans exposed to BPA, DEHP and NP

ABSTRACT: Gene expression analysis was conducted on the wildtype Caenorhabditis elegans exposed to bisphenol A (BPA), di(2-ethylhexyl) phthalate (DEHP) and nonylphenol (NP) using whole genome microarray. The microarray study was conducted in an ecotoxicological context, by investigating the response of global gene expression with that of classical toxicological endpoints, such as, mortality, growth, reproduction and development. Results provide insight into global transcription response of C.elegans to these endocrine disrupting chemicals exposure and also contribute to enhance the potential of C.elegans microarray in ecotoxicology (ecotoxicogenomics). key word : ecotoxicogenomics

Ecotoxicity of silver nanoparticles on the soil nematode Caenorhabditis elegans using functional ecotoxicogenomics

Differentially expressed genes from the microarray were selected for the quantitative analysis. The microarray study was conducted in an ecotoxicological context: the integration of gene expression with organism and population level endpoints (survival, growth, reproduction) was investigated, to test the ecotoxicological relevance of AgNPs-induced gene expression. Project description:Background: Toxicogenomics provides new opportunities for innovative and proactive approaches to chemical screening, risk assessment, and predictive toxicology. If applied to ecotoxicology, genomics tools could greatly enhance the ability to detect toxicants and understand the modes of toxicity in an environmental setting. However, few studies have yet to illustrate the potential of genomic techniques in ecotoxicology. Objective: Therefore, our objective was to demonstrate the potential utility of gene expression profiling in ecotoxicology using Daphnia magna, a standard aquatic ecotoxicity test organism. Methods: D. magna were exposed to copper, cadmium, and zinc at the 1/10 LC50 for 24 hours. Following each exposure, RNA was isolated, reverse transcribed, and the cDNA was hybridized to a 5000 clone cDNA microarray for D. magna. Differentially expressed cDNAs were sequenced and homology searches revealed each gene product's potential function. Real time PCR was used to verify the differential expression of several genes, and enzyme assays were used to assess the significance of these changes. Results: We identified distinct expression profiles in response to acute copper, cadmium, and zinc exposures and discovered specific biomarkers of exposure including two probable metallothioneins, and a ferritin mRNA with a functional IRE. The gene expression patterns support known mechanisms of metal toxicity and reveal novel modes of action including zinc inhibition of chitinase activity. Conclusions: Using a cDNA microarray for traditional ecotoxicology organism, D. magna, we have identified novel biomarkers of exposure and revealed possible modes of toxicity, providing experimental support for the utility of ecotoxicogenomics. Project description:Ecotoxicological studies with NPs are nearly all focused on testing one or few NPs. A more systematic approach to compare effects across many NMs with identified different but related characters/descriptors (e.g. different aspect-ratios or surface activities) are only sparsely reported especially in ecotoxicology. There is an urgent need to provide tools that allow us to fast develop an (eco)toxicological dataset containing comprehensive toxicity information relating to a diverse spectra of NPs characters, thus allowing the extrapolation of acquired information across NMs. NM libraries are an excellent tool for alternative testing and modeling. In the present study a custom-designed Fe doped TiO2 NM library was used covering of a wide spectrum of properties. Enchytraeus crypticus (Oligochateta) was used as test species since it is both environmentally relevant (as an important soil representative oligochaete and is also a standard species in soil ecotoxicology studies) and also a genomic model, with a high-throughput (HTP) genomic library. Effects were assessed at the organism (survival, reproduction) and gene level (differential gene expression, microarray).

Effect of nonylphenol on gene expression in Atlantic salmon liver, gill, hypothalamus, pituitary, and olfactory rosettes

Project description:Exposure to environmental contaminants like nonylphenol can disrupt smolt development and may be a contributing factor in salmon population declines. We used GRASP 16K cDNA microarrays to identify genes that are differentially expressed in the liver, gill, hypothalamus, pituitary, and olfactory rosettes of Atlantic salmon smolts treated with nonylphenol compared to control smolts. Nonylphenol treatment was confirmed using physiological assays: nonylphenol-treatment significantly decreased gill Na+,K+-ATPase activity and plasma cortisol and T3 levels. Microarray analyses were used to compare expression in nonylphenol-injected fish with expression in vehicle-injected fish: eight arrays each for liver, gill, olfactory rosettes, hypothalamus, and pituitary tissues. Total RNA was isolated from the tissues of eight nonylphenol-injected fish (six males and two females) and eight vehicle-injected fish (two males and six females) and reverse transcribed separately (not pooled); each slide represents a biological replicate. For each tissue, the eight arrays were balanced for dye: nonylphenol-injected fish were labeled with Alexa Fluor 555 and vehicle-injected fish were labeled with Alexa Fluor 647 on four slides, nonylphenol-injected fish were labeled with Alexa Fluor 647 and vehicle-injected fish were labeled with Alexa Fluor 555 on four slides. Liver, gill, hypothalamus, pituitary, and olfactory rosette tissues were analyzed separately.
