Identification of SRSF6 splicing regulatory map and its impact on diabetes susceptibility genes regulation

ABSTRACT: The downregulation of diabetes susceptibility gene GLIS3 contributes to pancreatic beta cell demise, at least in part, through downregulation of the splicing factor SRSF6. Here, we used individual-nucleotide UV crosslinking and immunoprecipitation (iCLIP) to map the RNA binding landscape of SRSF6 in pancreatic beta cells.

Cells grown in normal conditions were used as control (Normoxia, 21% oxygen). We performed iCLIP using anti-GFP antibodies and compared the binding pattern of SRSF6 between normoxia, 4h hypoxia and 24h hypoxia.

Overexpression of splicing factor SRSF6 in mouse skin

Project description:Many biological processes involve post-transcriptional regulation of gene expression by alternative pre-mRNA splicing. Here, we show that an alternative splicing factor SRSF6 affects tissue homeostasis of the skin. Identification of SRSF6 downstream regulatory genes and its impact on pleural fibrosis progression

Project description:Pleural fibrosis is defined as an excessive deposition of extracellular matrix that results in destruction of the normal pleural tissue architecture and compromised function. There is no effective medication for this disorder. Pleural mesothelial cells (PMCs/ Met-5A) play a key role in the process of pleural fibrosis. However, the detailed mechanisms are poorly understood. In our study, firstly SRSF6 protein has a major role in pleural fibrosis progression. To explore downstream of SRSF6, RNA sequencing was performed in Met-5A treated by bleomycin(0.2ug/ml) with SRSF6 siRNA or negtive control siRNA (siRNA NC). In conclusion, SRSF6 induced pleural fibrosis through a cluster pathway including SRSF6/WNT5A and SRSF6/SMAD1/5/9 signaling. SRSF6 is involved in pleural fibrosis, inhibition of SRSF6 is a novel strategy for treatment.

SRSF6 -regulated alternative splicing of genes involved in DNA repair is associated with prostate cancer progression

Project description:Alternative splicing (AS) occurs in nearly all human genes and abnormal AS has a close connection with cancer. SRSF6, a canonical member of the serine/arginine-rich (SR) protein family, has been characterized as an important regulator of AS. However, the role of SRSF6 in regulating AS in prostate cancer remains unclear. In this study we analyzed SRSF6 expression levels in 550 prostate cancer tissue samples available in The Cancer Genome Atlas database and found that expression of SRSF6 was upregulated in all prostate cancer stages we examined. To investigate the biological consequences of SRSF6 overexpression, we constructed an SRSF6-overexpressing cell model and utilized RNA-seq to explore transcriptional changes. We also selected two groups of prostate cancer tumor samples in which SRSF6 was differentially expressed to analyze potential SRSF6-regulated AS. We hypothesized that overexpression of SRSF6 in cancer cells would induce large-scale changes in transcriptional expression levels and AS. We found that the pattern of SRSF6-regulated AS in cancer cells was similar to that observed in clinical samples and was enriched in pathways like DNA repair, double-strand break repair via homologous recombination and others. We also validated SRSF6-regulated AS events using RT-qPCR. Our findings highlighted how SRSF6 could regulate DNA repair-related pathways via regulation of AS to influence cancer progression in both cancer cells and prostate cancer tumor samples. These results greatly expand our current understanding of the mechanisms underlying SRSF6-mediated gene regulation and suggests the potential use of SRSF6 as a cancer therapeutic target.

Glis3 regulates pancreatic beta cells development and maturation

Project description:Glis3 is expressed in pancreatic beta and PP cells. To identify down stream target genes of Glis3, we performed microarray analysis using pancreas total RNAs from 1 week-old WT and Glis3KO2 mice. insulin and pancreatic polypeptide (Ppy) was significantly decreased together with several other β cell markers, Glut2 and MafA by microarray analysis. Immunohistochemistry, QRT-PCR, and transmission electron microscopy indicated that postnatal Glis3KO2 pancreas still contains a large population of β cells that express Pdx-1, Nkx6.1, and Isl-1; however, insulin production and secretory granules were greatly reduced in these cells. In addition, chromogranin A (ChgA) and Urocortin 3, which are associated with mature β cells, was dramatically decreased in Glis3KO2 pancreas. These observations suggest that Glis3 plays a critical role in the maturation of pancreatic β cell phenotype.

T.F. Glis3: a novel critical player in the regulation of pancreatic beta-cell development and insulin gene expression

Project description:Glis3 mutant mice (Glis3zf/zf) die within the first week after birth due to overt diabetes, evidenced by hyperglycemia and hypoinsulinemia. Histopathological analysis showed that Glis3zf/zf mice develop a pancreatic phenotype with a dramatic loss of beta- (insulin) and delta- (somatostatin) cells contrasting a smaller relative loss of alpha- (glucagon), PP- (pancreatic polypeptide), and epsilon- (ghrelin) cells. Glis3zf/zf mice develop ductal cysts with decreased number of primary cilia, while the acini are not significantly affected. Gene expression profiling by microarray analysis demonstrated that the expression of terminal hormonal genes and several transcription factors important in endocrine development were significantly deregulated in Glis3zf/zf mice. During pancreatic development, Glis3 mRNA expression is induced during the secondary transition, a stage of cell lineage specification and extensive patterning. Changes in pancreatic development of Glis3zf/zf mice are noted during and after this stage. The population of pancreatic progenitors appears not to be greatly affected in Glis3zf/zf mice; however, the number of neurogenin 3 (Ngn3) positive, endocrine progenitors is significantly reduced. Our study indicates that Glis3 plays a key role in cell lineage specification, particularly the development of mature pancreatic beta-cells. In addition, we identified evidence that Glis3 regulates insulin gene expression through two Glis-binding sites in its proximal promoter indicating that Glis3 is a regulator of insulin gene expression.
