Project description:Five NPC cell lines (HONE1, CNE1, CNE2, 5-8F, and 6-10B) and an immortalized nasopharyngeal epithelial cell line NP69 were evaluated with 250K SNP arrays (Affymetrix) to detect the genome-wide DNA copy number alterations. The same type of data of 45 Chinese samples from the HapMap project was used as normal control. Data was analyzed using GCOS (GeneChip operating software, Ver.1.4), GTYPE (GeneChip Genotyping Analysis Software, Ver.4.1), and CNAT (chromosome copy number analysis tool, Ver.4.0, BRLMM algorithm).

Project description:To search for molecular processes underlying miR-18a-mediated oncogenesis in nasopharyngeal carcinoma(NPC) cells, whole-genome mRNA microarray analysis was performed using the HumanGene Expression 4 x 44k v2 Microarray Kit (Agilent Technologies) in 6-10B cells with stable miR-18a overexpression and in 5-8F cells with transient silencing of miR-18a by siRNA. Differentially expressed genes in miR-18a-overexpressing cell and differentially expressed genes in miR-18a knockdown cells were analyzed.

Expression profiling of nasopharyngeal carcinoma cell lines

Project description:To identify the deregulated genes which may involved in the carcinogenesis of nasopharyngeal carcinoma (NPC). To identify potential tumor suppressor genes / oncogenes of NPC, we have employed whole genome microarray expression profiling as a discovery platform to analyze two NPC cell lines, TW01 and HONE1. For the purpose to filter out differential genes may not related to carcinogenesis of NPC, a gene expression database for premalignant nasopharyngeal epithelial cells was obtained from NP460hTert cells, and we used the commercial universal human RNA (stratagene) as common reference for all microarray experiments. A total of 1272 genes were commonly up-regulated (fold change > 2) and 1810 genes were commonly down-regulated (fold change < 0.5) in TW01 and HONE1 cell lines, whereas not deregulated in NP460hTert cells.

Radioresistant subclone of nasopharyngeal carcinoma CNE2-IR cell line was cultured and produced according to the experienment schedule to undergo five rounds of sublethal dose of irradiation (11 Gy), and the parent cell line CNE2 cell line sensitive to radiotherapy as the control
