LncRNA expression alterations in mesothelial cells treated with exosomes

ABSTRACT: To investigate whether exosomes from ovarian cancer cell lines could change lncRNA expression in mesothelial cells, exosomes from ovarian cancer cells (SKOV3 cells, A2780 cells) were added to MeT-5A cells, PBS treatment was set as blank control. Total RNAs of MeT-5A cells treated with exosomes were extracted, and then lncRNA sequencing was performed.

Project description:To investigate whether the EVs from cancer cell lines could change gene expression in mesothelial cells, the EVs from five types of cells (ES-2 cells, A2780 cells, SKOV3 cells, HOSE1 cells and HOSE2 cells) were added to MeT-5A cells (Fig. 2a). All three selected cancer cell lines expressed metastatic phenotypes in mouse model (Fig. 1c), and EVs from HOSE2 cell line, established together with HOSE1 cell line, was also set as control. Total mRNAs were extracted by MeT-5A cells treated with EVs, and then microarray analysis was performed.

Project description:RNA-sequencing was performed to gain insight into the mechanism responsible for the mesenchymal-to-epithelial transition (MET) induced by loss of long non-coding RNA (lncRNA) DNM3OS in SKOV3 ovarian cancer cells. Following siRNA-mediated knockdown of DNM3OS or non-targeting control, RNA-sequencing was performed. This high-throughput data revealed knockdown of DNM3OS down-regulated the expression of genes and pathways known to induce EMT in ovarian cancer.

Project description:To further development of the effects of β-HCG in ovarian cancer SKOV3 cells，we have employed lncRNA and mRNA microarray as a discovery platform to identify lncRNA and mRNA expression in β-HCG overexpressing ovarian cancer cells. SKOV3 cells were transfected with lentiviral vector with eGFP, encoding β-HCG and negative control vector (LV- β-HCG and LV-CON,) by using polybrene. The dysregulation of β-HCG was confirmed by using RT-PCR. RNA was extracted and detected by a lncRNA and mRNA microarray in LV-β-HCG and LV-CON SKOV3 cells. The different expression of lncRNA and mRNA in LV-β-HCG and LV-CON SKOV3 cells was analyzed to explore the mechanism that β-HCG affect ovarain cancer cells.
