Effect of LLC1 tumor cell conditioned medium on RNA expression in mouse osteoblasts (MC3T3 cells)

ABSTRACT: MC3T3 osteoblasts were grown in culture medium in the presence of 10 mM beta-glycerophosphate and and 100 micro g per mL of ascorbic acid and 25% LLC1 conditioned medium or vehicle for 7 days. Cells were lysed an RNA was isolated.

Osteoblast differentiation experiment: BMP-2 induced osteogenesis in mouse MC3T3 cell line

Project description:MC3T3 cells clone 1b were grown in alpha-MEM with 10% fetal calf serum (FCS, Gibco), 1% penicilin/streptomycin (Pen/Strep) and 1% L-Glutamine (L-Glu). For differentiation experiment, cells were seeded on 6 cm dishes (3x105cells/dish in 5 ml medium) and grownt to confluence for 3 days at 37C / 5% CO2. Cells were then stimulated with osteogenic stimulus (10 mM beta-glycerophosphate (GP, Sigma), 50 µM ascorbic acid (AA, Wako) and 1 mg/ml BMP-2 (Novartis)), or with 10mM GP alone for 1 or 3 days. Control cells-day 0, medium. Experiment was done in triplicate. Total RNA was extracted, treated with DNAse, and purified according to the manufacturer’s protocol (RNeasy mini kit, QIAGEN). Expression data from control and Ascorbic Acid (AA) stimulated Mc-3T3-E1 osteoblasts

Project description:Despite advances in investigating functional aspects of osteoblast (OB) differentiation, especially studies on how bone proteins are deposited and mineralized, there has been little research on the intracellular trafficking of bone proteins during OB differentiation. Collagen synthesis and secretion is markedly upregulated upon Ascorbic Acid (AA) stimulation. Understanding the mechanism by which collagen is mobilized in specialized OB cells is important for both basic cell biology and diseases involving defects in bone secretion and deposition. RabGTPases are major regulators on protein trafficking throughout the cell. In this study, we identified the Rab GTPases that are upregulated during 5-day AA differentiation of OBs using microarray analysis, namely Rab1, Rab3d and Rab27b. We used microarrays to detail the global programme of gene expression underlying procollagen production and trafficking and identified up-regulated genes during this process. Control Mc3T3-E1 cells and 5 day Ascorbic Acid stimulated cells were processed for RNA extraction and hybridization on Affymetrix microarrays.
