RNAseq of GL261 tumor endothelial cells from wild type and Eltd1-/- mice

ABSTRACT: Eltd1 is widely expressed in endothelial cells. We have reported that the expression of Eltd1 is highly up-regulated in glioma vessels. Other studies have shown that Eltd1 is up-regulated in vessels in other types of tumors, and has been suggested as a vascular target for anti-angiogenesis therapy. However, the role of Eltd1 in vessels is largely unknown. We isolated endothelial cells from healthy brain tissue and GL261 tumor tissue, and analyzed the role of Eltd1 in tumor angiogenesis by analyzing the endothelial gene profile from wild type and Eltd1-/- mice.

Project description:Increased awareness for the role for angiocrine factors in stem cell biology, in general, has suggested a role for angiocrine factors in the maintenance of GSC stemness and also there are reciprocal bi-directional signals from endothelial cells (ECs) to glioma (Gl261) and vice versa. To extend, refine and elucidate perfusion-independent modes of GBM-EC communication (i.e., distinguishing contact dependent or contact independent routes), we carried-out the following experiments: ECs from umbilical veins tagged with RFP (HUVEC-RFP) were cultured with mouse glioma line Gl261 fluorescently labelled with GFP (Gl261-GFP) until reaching confluency by 72 h post culturing. Cells were than harvested, RNA extracted, and the combined co-culture transcriptomes were sequenced from a generated cDNA library without prior separation of the respective cell types (sample named as Co). The Co transcriptome was compared to that of an artificial mixtures of the two cell types (Gl261-GFP and HUVEC-RFP) in the exact cells ratio attained by the end of the co-cluttering aided by FACS-based determination of the GFP/RFP ratio (sample named as Sep). In this procedure advantage was taken on the different species from which GBM and ECs are derived and specialized bioinformatics tools allowing to unambiguously assign the transcripts to their respective GBM (mouse) or EC (human) origins, thereby circumventing confounding factors associated with cell purification. Considering that contact-dependent EC signaling is primarily regulated by canonical Notch pathway and that the Notch pathway was reported to play a role in self-renewal of GSCs, we also treated GBM-EC co-cultures with the Notch pathway inhibitor Gamma secretase inhibitor (GSI) (or with the DMSO vehicle alone) for 24 h before harvesting.

CircGLIS3 promotes high-grade glioma invasion via modulating Ezrin phosphorylation (microarray)

Project description:High-grade glioma is highly aggressive and malignant, resistant to combined therapies and easy to relapse. A better understanding of circRNA biological function in high-grade glioma might contribute to the therapeutic efficacy. Here, a circRNA merely up-regulated in high-grade glioma, circGLIS3 (hsa_circ_0002874, originating from exon 2 of GLIS3), was validated by microarray and qRT-PCR. Functional experiments uncovered that up-regulation of circGLIS3 promoted glioma cell migration and invasion, and showed aggressive characteristics in tumor-bearing mice. Fluorescence in situ hybridization, RNA pull-down, RNA immunoprecipitation and immunohistochemical staining showed that circGLIS3 could promoted Ezrin T567 phosphorylation. Further investigation showed that circGLIS3 could be excreted by glioma through exosomes and induced endothelial cells angiogenesis. This study indicates that circGLIS3 is up-regulated in high-grade glioma and contributes to the invasion and angiogenesis of glioma via promoting Ezrin T567 phosphorylation.

Identification of diverse tumor endothelial cell populations in malignant glioma

Project description:Background: Glioblastoma multiform is the most common and aggressive type of primary brain tumor, as most patients succumb to the disease less than two years after their diagnosis. Critically, studies demonstrate that glioma recruits surrounding blood vessels, while some work suggests that tumor stem cells themselves may directly differentiate into endothelial cells, yet the molecular and cellular dynamics of the endothelium in glioma are poorly characterized. The goal of this study was to establish clear molecular and morphological benchmarks for tumor associated vessels (TAVs) and tumor derived endothelial cells (TDECs) during GBM disease progression. Methods: Using In-Utero Electroporation and CRISPR/Cas9 genome engineering to generate a native, immunocompetent mouse model of glioma, we characterized vascular-tumor dynamics in three dimensions during tumor progression. We also employed bulk and single-cell RNA-Sequencing to elucidate the relationship between TAV and TDECs. We confirmed our findings in a patient derived orthotopic xenograft (PDOX) model. Results: Using a mouse model of glioma, we identified progressive alteration of vessel function and morphogenesis over time. We also showed that TDECs exist, and that these cells contribute to vessels within the tumor. Furthermore, transcriptional profiling demonstrates that both TAVs and TDECs are molecularly distinct, and that both populations feature extensive molecular heterogeneity. Finally, the distinct molecular signatures of these heterogenous populations are also present in human glioma. Conclusions: Our findings show extensive endothelial cell heterogeneity within the tumor and tumor microenvironment, and provide insights into the diverse cellular and molecular mechanisms that drive glioma vascularization and angiogenesis during tumorigenesis.

Transcriptomic heterogeneity of endothelial cells during devleopmental angiogenesis

Project description:Efficient angiogenesis, the formation of new blood vessels from pre-existing vessels, underlie tissue growth during development. Blood vessels in the mouse ear has been shown to dramatically expand at the neonatal stage to meet the needs of postnatal tissue growth. Yet, it is not clear at the transcriptome level how endothelial cells lining the innermost layer of blood vessels drive angiogenesis at this developmental stage. The highly heterogeneous endothelial cell types during angiogenesis render traditional bulk RNA-sequencing unsuitable. Therefore, we utilized single-cell RNA- and CITE-seq to profile transcriptomic heterogeneity of both blood and lymphatic endothelial cells during developmental angiogenesis in mouse ear.
