Transcriptomic study of the mechanism by Kai Yu Zhong Yu (KYZY) recipe to improve oocyte quality in the stressed mouse model

ABSTRACT: Female ICR mice were treated with stress or stress plus herbal medicine (KYZY decoction) in chronic unpredictable mild stress (CUMS) model. Fifteen isolated oocytes from each mouse were used for RNA-seq analysis. We used DESeq2 to identify differentially expressed genes (DEGs) between groups. DAVID and GO resources were used to perform gene enrichment analysis on the DEGs.

RNA sequencing analysis of gene expression in rat hippocampus exposed to chronic stress and albiforin treatment

Project description:We performed high-throughput profiling of gene expression in rat hippocampus in response to chronic unpredictable mild stress (CUMS) and albiflorin treatment. Total 415 differentially expressed genes (DEGs) were identified in rat hippocampus in response to albiflorin treatment compared with CUMS rats treated with saline (CUMS-Sal). We conducted the integrated metabolomics and transcriptomics analysis and found the correction of 16 biochemical pathways by albiflorin such as sphingolipids, phospholipids, tryptophan metabolism, fatty acid oxidation, and purine and pyrimidine metabolism. Our study provided deep insights into the understanding of the molecular mechanisms underlying the rapid antidepressant actions of albiflorin.

Molecular mechanism for stress-induced depression assessed by sequencing miRNA and mRNA in medial prefrontal cortex (miRNA)

Project description:Major depressive disorder is a common mood disorder. Chronic stressful life is presumably main etiology that leads to the neuron and synapse atrophies in the limbic system. However, the intermediate molecules from stress to neural atrophy remain elusive. Mice were treated by chronic unpredictable mild stress (CUMS) until demonstrating depression-like behaviors confirmed by the tests of sucrose preference, forced swimming and Y-maze. The sequencings of microRNA and mRNA from the medial prefrontal cortices were performed in CUMS-induced depression mice versus control mice to assess the molecular profiles of major depressive disorder. In the medial prefrontal cortices of depression-like mice, the levels of mRNAs that translated the proteins for the GABAergic synapses, dopaminergic synapses, myelination, synaptic vesicle cycle and neuronal growth were downregulated. miRNAs of regulating these mRNAs are upregulated. The deterioration of GABAergic and dopaminergic synapses as well as axonal growth is associated to CUMS-induced depression.

Project description:Two independent differential expression analyses were performed in mice subjected to chronic unpredictable mild stress (CUMS): one consisted of microglia (Cd11b+ cells) from the prefrontal cortex (PFC) and the other from the hippocampus (HC). After the CUMS protocol, minocycline, a wide spectrum antibiotic with anti-inflammatory properties, was delivered (CUMS_M group) in the drinking water/saccharine solution during 21 days. Transcriptomic profiling for a vehicle group (VEH), who spent 3 weeks in the cage with the same drinking solutions (Water/ saccharine 0.1%) and for a environmental enrichment (EE) group, that also spent 3 more weeks in the cage where enrichment materials were provided, were performed as well.

Stable isotope-resolved metabolomics reveals the abnormal brain glucose catabolism in depression based on chronic unpredictable mild stress rats

Project description:The severe harm of depression to human life has attracted great attention to neurologists, but its pathogenesis is extremely complicated and has not yet been fully elaborated. Here, we provided a new strategy for revealing the specific pathways of abnormal brain glucose catabolism in depression, which from the supply of energy substrates and the evaluation of mitochondrial structure and function. By using stable isotope-resolved metabolomics technique, we discovered the tricarboxylic acid cycle (TCA cycle) is blocked and the gluconeogenesis is abnormally activated in chronic unpredictable mild stress (CUMS) rats. In addition, our results showed an interesting phenomenon that the brain attempted to activate all possible metabolic enzymes in energy-producing pathways, but CUMS rats still exhibited a low TCA cycle activity due to impaired mitochondria. Depression caused mitochondrial structure and function impaired, and then led to abnormal brain glucose catabolism. The combination of the stable isotope-resolved metabolomics and mitochondrial structure and function analysis can accurately clarify the mechanism of depression. The mitochondrial pyruvate carrier and acetyl-CoA maybe the key targets for depression treatment. The strategy provides a unique insight for exploring the mechanism of depression, the discovery of new targets, and the development of ideal novel antidepressants.
