Project description:Intrauterine exposure to disturbed maternal glucose metabolism is associated with adverse consequences for the offspring. Hepatic disorders in affected offspring emerge in early development; thus, detection of disease biomarkers at an early stage may elucidate the underlying mechanisms of maternal hyperglycemia-induced metabolic disease and improve timely diagnosis and treatment strategies. To systematically study the molecular consequences of maternal hyperglycemia, we used data independent acquisition (DIA) proteomics and compared the molecular profiles of liver of three days old wild-type piglets born to a transgenic hyperglycemic pig model with those of wild-type piglets born to normoglycemic mothers.

Project description:Early in fetal development, maternal nutritional environments are expected to induce changes in epigenetic regulation in offspring.In the present study, we integrated methylome and transcriptome data to identify potential salt-sensitive hypertension genes using the kidneys of pups exposed to a low-protein diet throughout their fetal life. We aimed to uncover how specific candidate marker genes are modified over time and under different nutritional interventions at an early age. Using the kidneys of pups exposed to a low-protein diet (LP) compared to those exposed to a control diet (CN) during fetal life, we searched for candidate genes by integrating data from both methylome and transcriptome profiling.Transcriptome analyses were used to identify genes with differential expression.

Project description:Intrauterine growth restriction (IUGR) increases the risk of developing type 2 diabetes during adulthood. At day 18 of gestation, we used bilatual uterine artery ligation (BUAL) to restrict nutrient supply to developing rat fetuses to produce IUGR pups. At 2 and 10 weeks of age pancreatic islets were isolated and total RNA extracted using Trizol. Samples with RNA integrity numbers greater than 7 were used to generate libraries
