Single cell analysis of PVNS synovium from human

ABSTRACT: Pseudotime analysis, cytotrace, velocity was used to determine the source of subpopulation. To compare the difference between PVNS and steady state synovium, we conduct collaborated analysis containing both PVNS and OA synovium. Qusage analysis reveal the difference of the subsets between them. We for the first time using single cell analysis delineating PVNS cell atlas and have deepen understanding toward PVNS Presumed function and source of certain subpopulation might be the future target that worth trying. We also locate major subpopulation that is responsible for its unremitting inflammation, angiogenesis and invasiveness.

Single cell RNA-sequencing of the human synovium in inflammatory arthritis

Project description:The study presents: - an optimized synovium dissociation protocol for single cell RNA-sequencing studies of the human synovium. The protocol enables the isolation of high yield of viable synovial cells from prospectively collected fresh synovial biopsies from patients with inflammatory arthritis with a minimal sample droupout. The protocol is derived from the method for dissociation of cryopreserved synovia published by Donlin and colleagues (Arthritis Res. Ther. 2019). - a reference single-cell atlas of fresh human synovium in inflammatory arthritis, comprising more than 100´000 unsorted synovial scRNA-seq profiles from 27 freshly dissociated synovia of patients with different types of inflammatory arthritis. The synovial cells segregate into ten lymphoid, 14 myeloid and 17 stromal synovial cell populations and subpopulations, including synovial neutrophils, representing broadly representing the cellular heterogeneity and composition of the human synovium in inflammatory arthritis.

Project description:36 Yucatan minipigs underwent anterior cruciate ligament (ACL) transection and were randomly assigned in equal numbers to no further treatment, reconstruction or ligament repair. Peri-meniscal synovium was harvested at 1 and 4 weeks post-operatively and histology and RNA-sequencing performed. The generated data served to identify the molecular pathophysiology present in inflamed synovium during the early development of post-traumatic osteoarthritis (PTOA), as well as differences between surgical treatments.

Transcription profiling of mouse knee joint synovium from different grades of inflammation in mouse collagen induced arthritis (CIA)

Project description:Knee joint synovium was used for gene expression analysis of mouse collagen induced arthritis (CIA). Synovium was prepared at day 30 after initial sensitization from: healthy controls, CIA animals with no, with mild, with moderate, or with severe joint inflammation. Each sample group is represented by three replicates, each consisting of tissue collected from three to four animals. Experiment Overall Design: The data set consists of 15 samples: five groups with three replicates each. One sample group is from healthy controls, the other groups are from CIA animals with different degress of joint inflammation.

Project description:After the injection of human synovial mesenchymal stem cells into the rat (Lewis strain) knee joint, synovium was investigated using species specific microarray analyses of human and rat transcripts to investigate the gene expression change after injection. Human synovial mesenchymal stem cells were injected into the rat anterior cruciate ligament transected knee. After harvesting the synovium and extraction of RNA, species specific microarray analyses were performed uisng Affymetrix microarrays to investigate the gene expression change of human and rat transcripts after injection.

Unilateral mastication induced alternations of microRNAs expression profiling in synovium of rat temporomandibular joint

Project description:The present study investigated the alterations of miRNA expression profiling in synovium of temporomandibular joint(TMJ) in a rat model of experimental unilateral mastication. The synovium tissue of TMJ condyles from rats with 4- and 8-weeks unilateral mastication were harvested to extract miRNA for profiling holistic expression alternations. A total of 20 differentially expressed miRNAs were identified. Except for 3 miRNAs with uncertain function, those miRNAs are associated with cell proliferation, invasion, and osteoblast differentiation process.
