Gene Expression in the developing hindstomach and forestomach of wild type and GATA4 conditional knock out and knock in embryonic mice

ABSTRACT: Gene expression was compared between wild type forestomach and hindstomach epithelial cells at embryonic day E14.5. Gene expression was compared between GATA4 knock out hindstomach epithelial cells and wild type hindstomach epithelial cells at embryonic day E14.5. Gene expression was compared between GATA4 knock in forestomach epithelial cells and wild type forestomach epithelial cells at embryonic day E14.5.

Aha-enrichment experiments consisted of in vivo protein labeling with azidohomoalanine (Aha) followed by tissue fractionation of the forelimbs and enrichment of labeled ECM proteins from the final IN pellet ('enriched'). 'Unenriched samples', or the background from which newly synthesized proteins were enriched from, were also analyzed via LC-MS/MS.

Project description:Wnt-4 signaling is critical for embryonic female sexual development. When Wnt-4 gene is deleted during embryonic development, the knock-out females present a partial sex reversal. We used microarrays to detail the global programme of gene expression underlying sexual development and identified distinct classes of up and down-regulated genes during this process. Mouse embryonic gonad from wild type and Wnt-4 knock-out mice (male and female) were selected at two stages of sex determination for RNA extraction and hybridization on Affymetrix microarrays. We sought to obtain homogeneous populations of embryonic gonad at each developmental stage in order to increase the temporal resolution of expression profiles. To that end, we hand-selected embryos according to morphological criteria at two time-points: E 12.5 and E14.5.

Sub-chronic forestomach transcriptional response in adult male MutaTMMouse following oral exposure to benzo(a)pyrene

Project description:Benzo(a)pyrene is a well-established human carcinogen in humans and rodents. In the present study, we sought to determine the dose- and time-dependent changes in gene expression upon oral exposure to benzo(a)pyrene. Adult male MutaTMMouse were exposed to three doses of benzo(a)pyrene or vehicle control (olive oil) for 28 days and sacrificed three days after the final exposure. This experiment examined the forestomach transcriptional response of male mice exposed to BaP for 28 days at three different doses, including D1 (25 mg/kg BW/day), D2 (50 mg/kg BW/day), and D3 (75 mg/kg BW/day) and vehicle control. Each dose group was examined 72 hours following the final exposure. Each dose group and time point had 4-5 biological replicates. There were a total 17 samples (arrays) included in the final analysis using a two-colour reference design.
