ALI1 interacts with N-acyl homoserine lactone and is involved in the perception of the quorum sensing molecule

ABSTRACT: In this report, we investigated oxo-C14-HSL-interacting proteins in plants. We propose the AHL-Interacting Protein 1 (ALI1) (previously known as AtGalk2) as interacting protein for oxo-C14-HSL in Arabidopsis. We present here several levels of confirmation and suggest that ALI1 is required for the perception of oxo-C14-HSL in Arabidopsis. This is the first direct evidence of a plant protein interacting with a bacterial quorum sensing molecule.

Project description:In order to gain coherent insights into plant responses we performed transcriptional analysis of Arabidopsis seedling after treatment with three different AHLs: N-hexanoyl-L-homoserine lactone (C6-HSL), N-3-oxo-decanoyl-L-homoserine lactone (oxo-C10-HSL), and N-3-oxo-tetradecanoyl-L-homoserine lactone (oxo-C14-HSL). Furthermore, we analyzed the transcriptome of oxo-C14-HSL pretreated plants after a secondary challenge with 100 nM flg22 for 2 and 24 hours after treatment. Gene expression in Arabidopsis thaliana seedlings were measured after a 3-days-pretreatment with 6 ÂµM of three different N-acyl homoserine lactones in comparison to plants treated with the coresponding volume of acetone (solvent control). Plants pretreated with oxo-C14-HSL were in addition treated with 100nM of flg22 to induced defense response. Three independent experiments were performed (replicates).

A New Class of Homoserine Lactone Quorum Sensing Signals

Project description:Quorum sensing is a term used to describe cell-to-cell communication that allows cell density-dependent gene expression. Many Gram-negative bacteria use acyl-homoserine lactone (acyl-HSL) synthases to generate fatty acyl-HSL quorum sensing signals, which function with signal receptors to control expression of specific genes. The fatty acyl group is derived from fatty acid biosynthesis and provides signal specificity, but the variety of signals is limited. We have discovered that the photosynthetic bacterium Rhodopseudomonas palustris uses an acyl-HSL synthase to produce p-coumaroyl-HSL by using environmental p-coumaric acid rather than fatty acids from cellular pools. The bacterium has a signal receptor with homology to fatty acyl-HSL receptors that responds to p-coumaroyl-HSL to regulate global gene expression. We also found that p-coumaroyl-HSL is made by other bacteria including Bradyrhizobium BTAi1 and Silicibacter pomeroyi DSS-3. This discovery extends the range of possibilities for acyl-HSL quorum sensing and raises fundamental questions about quorum sensing within the context of environmental signaling. Alterations in the extracellular proteome of human pathogen Pseudomonas aeruginosa in response to natural and modified N-acyl-L-homoserine lactones

Project description:The common opportunistic human pathogen Pseudomonas aeruginosa employs quorum sensing QS to regulate a large set of genes involved in virulence and host-pathogen interactions. The Las circuit positioned on the top of the QS hierarchy in P. aeruginosa, makes use of N-acyl-L-homoserine lactones (AHL) as signal molecules, like N-3-oxo-dodecanoyl-L-homoserine lactone (3O-C12-HSL). Here, a quantitative proteomic approach was used to study the effect of natural 3O-C12-HSL and four AHL analogues on the expression and excretion of QS-regulated extracellular proteins. Treatment with AHL compounds resulted in significant difference in appearance of the 3O-C12-HSL-responsive reference proteins related to QS communication and virulence, i.e., a distinct activity as QS modulators. In summary, 3O-C12-HSL has a profound effect on extracellular proteome involved in the pathogenicity of P. aeruginosa, and the four AHL analogues have achieved a distinct inhibitory effect on the extracellular proteome.

The activity of Rhodopseudomonas palustris p-coumaroyl-homoserine lactone-responsive transcription factor RpaR

Project description:The Rhodopseudomonas palustris transcriptional regulator RpaR responds to the RpaI-synthesized quorum-sensing signal p-coumaroyl-homoserine lactone (pC-HSL). Other characterized RpaR homologs respond to fatty acyl-HSLs. We show here that RpaR functions as a transcriptional activator, which binds directly to the rpaI promoter. We developed an RNAseq method that does not require a ribosome depletion step to define a set of transcripts regulated by pC-HSL and RpaR. The transcripts include several non-coding RNAs. A footprint analysis showed that purified His-tagged RpaR (His6-RpaR) binds to an inverted repeat element centered 48.5 base pairs upstream of the rpaI transcript start site, which we mapped by S1 nuclease protection and primer extension analyses. Although pC-HSL-RpaR bound to rpaI promoter DNA it did not bind to promoter regions of a number of RpaR-regulated genes not in the rpaI operon. This indicates that RpaR-control of these other genes is indirect. Because the RNAseq analysis allowed us to track transcript strand specificity we discovered that there is pC-HSL-RpaR-activated antisense transcription of rpaR. These data raise the possibility that this antisense RNA or other RpaR-activated non-coding RNAs mediate the indirect activation of genes in the RpaR-controlled regulon. The Rhodopseudomonas palustris p-Coumaroyl-HSL-RpaR regulon as defined by Not-so-random RNASeq. Three sets of comparisons: wt + vs - pC-HSL; wt vs rpaR mutant both + pC-HSL; rpaR + vs - pC-HSL.

Heterologous acyl-homoserine lactone synthase expression in Pseudomonas putida

Project description:Many bacteria harbor an incomplete quorum sensing system, wherein they possess LuxR homologues without the quorum sensing acyl-homoserine lactone (AHL) synthase, which is encoded by a luxI homolog. An artificial AHL-producing plasmid was constructed using a cviI gene encoding for C6-AHL (HHL) synthase from Chromobacterium violaceum and was introduced successfully into both wild-type and the ppoR (a luxR homolog) mutant. Our data provides evidence to suggest that the PpoR-HHL complex, but neither PpoR nor HHL alone, could attenuate growth, antibiotic resistance, and biofilm formation ability. In contrast, swimming motility, siderophore production, and indole degradation were enhanced by PpoR-HHL. The addition of exogenous indole increased biofilm formation and reduce swimming motility. Interestingly, indole proved ineffective in the presence of PpoR-HHL, thereby suggesting that the PpoR-HHL complex masks the effects of indole. Our data was supported by transcriptome analyses showing that the presence of the plasmid-encoded AHL synthase altered the expression of many genes on the chromosome in strain KT2440. Our results showed that heterologous luxI expression occurring via horizontal gene transfer can regulate a broad range of specific target genes, resulting in alterations of the phenotype and physiology of host cells.
