ScRNAseq of TAZ-CAMTA1 expressing endothelial cells in vivo

ABSTRACT: TAZ-CAMTA1 is a defining genetic aberration in the vascular cancer epithelioid hemangioendothelioma. We generated a novel transgenic mouse that expresses TAZ-CAMTA1 in endothelial cells. These mice develop vascular tumors in the lung and die by average on day 40. We isolated GFP + endothelial cells from three five-week old TRE-TAZ-CAMTA1;Cdh5-tTA;TRE-GFP mice and subjected these endothelial cells to scRNA-seq via the 10X genomics platform

Project description:The WWTR1(TAZ)-CAMTA1 and YAP1-TFE3 gene fusions are disease defining gene alterations for epithelioid hemangioendothelioma, a vascular cancer. The resultant fusion proteins fuse the C terminus of CAMTA1 and TFE3 in frame to the N terminus of TAZ and YAP, respectively. An unbiased BioID-mass spectrometry/RNAi screen identified YEATS2 and ZZZ3 as components of the Ada2a-containing histone acetyltransferase complex and key interactors of both TAZ-CAMTA1 and YAP-TFE3. An integrative NGS approach including RNA-Seq, ChIP-Seq, and ATAC-Seq showed TAZ-CAMTA1 and YAP-TFE3 transcriptional programs overlap with TAZ and YAP but also drive expression of a novel transcriptome.

An integrative NGS approach validating the chromatin occupancy of TAZ-CAMTA1 and YAP-TFE3 with distal regulatory elements, such as enhancer regions marked by the presence of H3K27ac marks. The altered transcriptional program of the fusion proteins owing to altered DNA binding as well as shifts in the chromatin landscape.

Whole Transcriptome analysis of Aurintricarboxylic acid treated NIH3T3 cells

Project description:Aurintricarboxylic acid (ATA) was identified as a potent inhibitor of the TAZ/TEAD transcriptional complex via a Alpha-LISA screen. We have previously demonstrated that a hyperfunctional form of TAZ, TAZ-CAMTA1, is sufficient to induce the development of a rare vascular sarcoma, Epithelioid Hemangioendothelioma. We have demonstrated that TAZ-CAMTA1 can similarly transform NIH3T3 cells to yield anchorage independent growth in vitro and this action requires the formation of a TAZ-CAMTA1/TEAD complex. To identify if ATA is able to inhibit the transcriptional action TAZ-CAMTA1/TEAD complex, whole transcriptomic analysis was performed on TAZ-CAMTA1 stably transfected NIH3T3 cells with or without ATA.

scRNA-Seq of Cdkn2a mutant murine Epithelioid Hemangioendothelioma tumors

Project description:Epithelioid hemangioendothelioma (EHE) is a rare vascular sarcoma that is associated with a TAZ-CAMTA1 fusion in greater than 90% of cases and loss of CDKN2A in human EHE tumors is associated with late stage disease. Murine EHE tumors were created by conditionally expressing TAZ-CAMTA1 and conditionally knocking out Cdkn2a within vascular endothelium. Resultant tumors were used for single cell RNA sequencing and compared against age matched wild type mice tissue.

Cell lines were generated from WWTR1-CAMTA1 positive murine EHE tumors which also harbor a loss of CDKN2A. RNA sequencing was performed on cell lines.
