MRNA expression profiling A549 lung cancer cells following PPARGC1A suppression

ABSTRACT: PPARGC1A oppositely regulates cancer metastasis in melanoma, breast, and pancreatic cancer; however, little is known about its impact on lung cancer metastasis. We generated gene-expression profile of control and PPARGC1A suppressed A549 cells, a lung adenocarcinoma cell line that expresses moderate levels of PPARGC1A to investigate the role of this gene in lung cancer metastasis.

Impact of knocking out Ppargc1a in intestinal stem cells on mucosal homeostasis

Project description:We report the application of single cell RNAseq assay in examining the effect of Ppargc1a gene expression in intestinal stem cells on mucosal homeostasis. Lgr5EGFPcreERT2 mice were crossed with Ppargc1a F/+ or F/F to selectively target gene expression in Lgr5 expressing intestinal stem cells. Mice were fed with control diet AIN76A from weaning for 3 months, and deletion of Ppargc1a gene was conducted via Tamoxifen injection. Mice were sacrificed 3 days post tamoxifen injection.

Project description:To determine genes regulated by the transcriptional co-activator PPARGC1A in OAC cells and whether PPARGC1A activity is altered by lapatinib we performed RNA-seq on OE19 cells treated with non-targeting siRNA or siRNA targeting PPARGC1A. 24 hours post-transfection, cells were also treated with either DMSO or 500 nM lapatinib for 24 hours.

GLIDR-mediated Regulation of Tumor Malignancy and Cisplatin Resistance in Non-Small Cell Lung Cancer via the miR-342-5p/PPARGC1A Axis

Project description:LncRNAs play a crucial role in regulating the progression of NSCLC, making them potential targets for cancer diagnosis and treatment. Therefore, identifying new lncRNAs as therapeutic target and comprehending their underlying regulatory mechanisms are crucial for treating NSCLC. In this study, we found that glioblastoma down-regulated RNA (GLIDR) was increased in non-response NSCLC patients with the primary treatment, and GLIDR was also highly expressed in NSCLC patients. We used bioinformatic analysis and luciferase assays to identify that microRNA-342-5p (miR-342-5p) directly targets GLIDR. MiR-342-5p overexpression inhibits NSCLC cell proliferation, migration, and invasion, whereas miR-342-5p inhibition promotes NSCLC malignancy, which can be rescued by suppressing GLIDR. Peroxisome proliferator-activated receptor-gamma coactivator-1alpha (PPARGC1A) was identified as a downstream target of miR-342-5p. PPARGC1A inhibition significantly suppresses NSCLC malignancy, whereas overexpression promotes it. The results also revealed that GLIDR overexpression reduced PPARGC1A inhibition by miR-342-5p, and increased PPARGC1A expression reversed the inhibition of NSCLC malignancies caused by decreased GLIDR. To the best of our knowledge, we are the first to discover that GLIDR promotes NSCLC progression by sponging miR-342-5p to regulate PPARGC1A expression, presenting that GLIDR act as a potential therapeutic target in NSCLC.

Genome-wide maps of the total and lysine methylated enriched pools of peroxisome proliferating activated receptor gamma (pparg) co-activator 1 alpha (ppargc1a) in Hepa 1-6 cells

Project description:We report the application of single nucleotide sequencing technology for high-throughput profiling of ppargc1a in mouse hepatome cells (Hepa1-6). By obtaining 1.2 billion bases of sequence from chromatin immunoprecipitated DNA, we generated genome-wide maps of ppargc1a sin mouse hepatoma cells. We identify novel non- coding sites of ppargc1a occupation in hepatic cells for Sirt5, Idh3b, Pfkl, and Hmox2 strongly corresponds with the enrichment of enhancer -associated RNAs. Examination of the genomic disttribution of the total pool of ppargc1a and the lysine methylated (K779me) enriched pool of ppargc1a in mouse Hepa1-6 hepatoma cells.

ADAM9 Up-regulates N-Cadherin via MiR-218 Suppression in Lung Adenocarcinoma Cells [mRNA]

Project description:Lung cancer is the leading cause of cancer death worldwide. Brain metastasis is a major cause of morbidity and mortality in lung cancer. CDH2 (N-cadherin, a mesenchymal marker in epithelial-mesenchymal transition) and ADAM9 (a member of type I transmembrane proteins) have been reported relating to lung cancer brain metastasis, however, it is still not clear whether any interaction between them to mediate lung cancer brain metastasis. Since microRNAs were discovered to regulate many biological functions and disease processes (e.g., cancer) by down-regulating their target genes, microRNA microarrays were used to identify ADAM9 regulated miRNAs that target CDH2 in aggressive lung cancer cells. Luciferase assays and immunoblotting proved that CDH2 was a target gene of miR-218. The expression of miR-218 was generated from pri-mir-218-1, located in SLIT2, in low invasive lung adenocarcinoma while it was inhibited in aggressive lung adenocarcinoma. Down-regulation of ADAM9 could up-regulate SLIT2 and miR-218, thus down-regulate CDH2 expression. This study elucidated the mechanism of ADAM9 activating CDH2 may be due to release the inhibition of miR-218 on CDH2 in lung adenocarcinoma. For each of the cell lines bm#2, bm#7, and F4, one microarray was analyzed.
