ABSTRACT: C57BL/6 mice were exposed by oropharyngeal aspiration to 28 nanomaterials including different surface functionalizations on 4 concecutive days with the dose of 10ug/day. Total RNA was collected from the mice lung biopsies after sacrificing.

Inhalation and oropharyngeal aspiration exposure to rod-like carbon nanotubes induces highly similar airway inflammation and biological pathways in mouse lungs

Project description:Inhalation and low-dose aspiration elicit highly similar pulmonary inflammation which was confirmed by activation of essentially similar biological pathways and even the same genes. Present results provide evidence that oropharyngeal aspiration is valid approach for hazard assessment of nanomaterials and proper alternative method for inhalation exposure.

MRNA-Integrative multi-omics survey of effects carbon-based engineered nanomaterials on lung derived cell-lines

Project description:In this study we exposed three human lung derived cell lines to sublethal dosages of nanomaterials for a limited amount of time. For the first time, we assessed the simultaneous effects of nanomaterials exposure on three distinct molecular layers along with their interactions in the determination of the MOA of 10 carbon based nanomaterials. By performing an integrative analysis we provide here a complete picture of the interaction between regulatory factors (DNA methylation and miRNAs) and mRNA deregulation subsequent to exposing different cellular systems to different nanomaterials.

Multi-omics toxicity profiling of engineered nanomaterials

Project description:This study aims to implement methods for toxicological profiling of engineered nanomaterials using toxicogenomics, proteomics, and metabolomics along with computational analyses. For all three omics layers the human cell lines A549 (lung epithelial cells) and THP1 (monocytes) were separately exposed to the nanomaterials TiO2 NM104, MWNCT NM401, and Ag NM300k. Proteomics and metabolomics samples have been performed as biological triplicates and were taken after 0, 6, 12, 24, and 48 hours. To assess ecotoxic effects C. elegans worms were grown in soil treated with NM300k. Ecotox samples were taken only at 0 and 24 hours. Integrating all three omics layers will enable the identification of (novel) ENM specific modes of action (MoA).
