Leaf margins of indeterminate Arabidopsis leaves

Project description:Leaves are flat determinate organs derived from indeterminate shoot apical meristems. The presence of a specific leaf meristem is debated, as anatomical features typical of meristems are not present in leaves. Here we demonstrate that multiple NGATHA (NGA) and CINCINNATA-class-TCP (CIN-TCP) transcription factors act redundantly to suppress activity of a leaf margin meristem in Arabidopsis thaliana, and that their absence confers persistent marginal growth of leaves, cotyledons and floral organs. The marginal meristem is activated by the juxtaposition of adaxial and abaxial domains and maintained by WOX homeobox transcription factors, but other margin elaboration genes are dispensable for its maintenance. This genetic framework parallels the morphogenetic program of shoot apical meristems and may represent a relic from an ancestral shoot system from which seed plant leaves evolved.

Analysis of wound-induced transcripts in poplar leaf disks versus whole leaves

Project description:Expression of defense response-related transcripts could be affected upon leaf wounding such as after excision of leaf disks. In this experiment, transcripts were isolated from whole leaves and 5-cm2 excised leaf disks samples from interaction assays between leaves of the 'Beaupré' poplar cultivar (Poplus trichocarpa x Poplus deltoides) and virulent (98AG31, compatible interaction) and avirulent (93ID6, incompatible interaction) urediniospores of Melampsora larici-populina, the fungus responsible for the foliar rust disease in poplar. For each type of interaction, three samples were collected in a time-course series (18, 21 and 24 hpi for strain 93ID6, and 18, 24 and 48 hpi for strain 98AG31). Both leaf disks and whole leaves were inoculated in strictly similar conditions, so differential transcript expression is likely attributable to the wounding effect in leaf disks. The aim of this study is to identify genes that are induced by wounding in leaf disks compared with whole leaves. For this purpose, cDNA templates were used for hybridization on poplar oligoarrays.

Gene expression in leaves of grapevine plants under abiotic stress conditions: heat and water

Project description:Experimental research on the effects of abiotic stress over grapevine has mainly focused on water shortage. The adaptation of plants to stress is a complex response triggered by cascades of molecular networks involved in stress perception, signal transduction, and the expression of specific stress-related genes and metabolites. Approaches such as array-based transcript profiling allow assessing the expression of thousands of genes in control and stress tissues. The variety AragonÃªs, used in wine production, was subjected to controlled individual abiotic stresses, WS and HS. Physiological effects were confirmed by measuring photosynthesis light curves at ambient CO2 and stepwise increasing irradiances. To assess physiological effects of stress treatments, light responses (A/I) curves were measured on the third fully expanded leaf from four plants per treatment and in the control, immediately after heat stress, and when Î¨w was -0.9 MPa in water stress.

Glycosyltransferase-like RSE1 negatively regulates leaf senescence by modulating salicylic acid signaling in Arabidopsis

Project description:Leaf senescence is a developmental process designed for nutrient recycling and relocation to maximize growth competence and reproductive capacity of plants. Thus, plants integrate developmental and environmental signals to precisely control senescence. However, it remains largely elusive as to how plants coordinate genetic, epigenetic, and metabolic pathways for the regulation of senescence. To genetically dissect the complex regulatory mechanism underlying leaf senescence, we identified an early leaf senescence mutant, rse1. RSE1 encodes a putative glycosyltranferase. Loss-of-function mutations in RSE1 resulted in precocious leaf yellowing and up-regulation of senescence maker genes, indicating enhanced leaf senescence. Transcriptome analysis revealed that salicylic acid (SA) and defense signaling cascades were activated in rse1 prior to the onset of leaf senescence. In agreement with the phenotypes, we found that SA accumulation was significantly increased in rse1. We also discovered that the rse1 phenotypes are dependent on SA-INDUCTION DEFICIENT 2 (SID2), indicating a role of SA in accelerated leaf senescence in rse1. Furthermore, RSE1 protein was localized to the cell walls, and rse1 displayed increased glucose contents in the cell walls, implying a possible link between the cell walls and RSE1 function. Together, we show that RSE1 negatively modulate leaf senescence through an SID2-dependent SA signaling pathway.
