Project description:Pathogen secreted pathogen-associated molecular patterns (PAMPs) play a key role in PAMPs triggered immunity (PTI) in plant for the recognition of pathogens. In rice, fewer PAMPs and their pattern recognition receptors (PRRs) have been characterized during rice-Magnaporthe oryzae interaction. Particularly, recent study was identified M. oryzae snodprot1 homolog known as MSP1, however, the molecular mechanism of MSP1 induced PTI is currently elusive. Therefore, we were generated the MSP1 overexpressed transgenic rice with their subcellular localization in the apoplastic (with signal sequence) and cytoplasmic (without signal sequence) regions, respectively, to examine the functional role of MSP1 in rice. Here, we employed a tandem mass tag (TMT)-based quantitative membrane proteomic analysis to decipher the potential interaction PRRs and MSP1-induced downstream signaling. This approach led to the identification of 8,033 proteins and sequential statistical analysis were identified 2,226 differentially modulated proteins. Of these, 20 plasma membrane localized receptor like kinases (RLKs) with the increased abundance in response to MSP1. Moreover, activation of proteins related to the protein degradation and modification, calcium signaling, transcription factor, redox, and MAPK signaling were characterized. Taken together, our results indicated that potential PRR candidates involved in response to blast disease and thus suggesting the overview mechanism of the MSP1-induced signaling in rice leaves.

Deciphering the differences in the complex signaling events that lead to pathogen recognition and initiation of the correct response remains challenging. Here we report the discovery of temporal changes in the protein signaling components involved in innate immunity. Using an integrated strategy combining unbiased proteomics, transcriptomics and macrophage stimulations with three different PAMPs, we identified differences in signaling between individual TLRs and revealed specifics of pathway regulation at the protein level.

We have identified proteobacterial translation initiation factor 1 (IF1) as an immunogenic pattern that triggers PTI in Arabidopsis thaliana and some related Brassicaceae species.

In addition to forming macrophages and dendritic cells, monocytes in adult peripheral blood retain the ability to develop into osteoclasts, mature bone-resorbing cells. The extensive morphological and functional transformations that occur during osteoclast differentiation require substantial reprogramming of gene and protein expression. Here we employ -omic-scale technologies to examine in detail the molecular changes at discrete developmental stages in this process (precursor cells, intermediate osteoclasts, and multinuclear osteoclasts), quantitatively comparing their transcriptomes and proteomes.

Project description:Plants have a wide variety of ways to defend against pathogens. A commonly used model of the plant immune system divides it into a general response triggered by pathogen associated molecular patterns (PAMPs) and a specific response triggered by effectors. The first type of response is known as PAMP triggered immunity (PTI) and the second as effector-triggered immunity (ETI). We have performed a proteomical analysis of one PTI and two ETI models in potato and compared their effect on protein expression.

Project description:A first line of defense against pathogen infections is the recognition of pathogen-associated molecular patterns (PAMPs), leading to PAMP-triggered immunity (PTI). MicroRNAs (miRNAs) are primarily known as central regulators of plant development, but a few have also been connected to immunity. We have found that several fungal pathogens lead to a reduction in miR396 levels, suggesting that miR396 are negative regulators of downstream defense responses. In agreement with such as scenario, constitutive attenuation of miR396 activity enhances resistance to infection by fungal pathogens, while increased miR396 activity reduces pathogen resistance. We conclude that constitutive reduction of miR396 levels confer a primed state for enhanced defense reactions

Project description:Plants deploy cell surface and intracellular leucine rich-repeat domain (LRR) immune receptors to detect pathogens. LRR receptor kinases (LRR-RKs) and LRR receptor proteins (LRR-RPs) recognise microbe-derived molecules to elicit pattern-triggered immunity (PTI), whereas nucleotide-binding LRR (NLR) proteins detect microbial effectors inside cells to confer effector-triggered immunity (ETI). Although PTI and ETI are initiated in different host cell compartments, they rely on the transcriptional activation of similar sets of genes, suggesting pathway convergence upstream of nuclear events. We report that PTI triggered by Arabidopsis LRR-RP (RLP23) requires signalling-competent dimers of the lipase-like proteins EDS1 and PAD4, and ADR1-family helper NLRs, which are all components of ETI. The cell surface LRR-RK SOBIR1 links RLP23 with EDS1, PAD4 and ADR1 proteins, suggesting formation of constitutive supramolecular complexes containing PTI receptors and transducers at the inner side of the plasma membrane.

Project description:PAMP-triggered immunity (PTI) is the first line of plant defense against invading organisms. Initiated through the perception of conserved pathogen-associated molecular patterns (PAMPs), such as lipopolysaccharide or flagellin, PTI can provide early protection against a broad range of pathogens. Active suppression of PTI by microbial effector proteins, particularly those secreted by the Type III secretion system (T3SS), is a well-known strategy employed by bacterial plant pathogens that enables them to subvert PTI and successfully colonize their hosts. In this study, we demonstrate that the xylem-limited bacterium, Xylella fastidiosa, which lacks a T3SS, utilizes an alternative strategy to delay elicitation of innate immune responses. By decorating its LPS PAMP molecule with a high molecular weight O antigen, this bacterium physically masks itself from early recognition by the grapevine innate immune system. We have elucidated the chemical structure of the O antigen and found that it is primarily an α1,2-linked rhamnan polymer. X. fastidiosa cells lacking O antigen elicited hallmarks of PTI such as ROS production, specifically in the plant xylem tissue compartment, which is comprised primarily of non-living cells. By coupling histological and genome-wide transcriptional profiling, we demonstrate that X. fastidiosa lacking its O antigen shield activates defense-related genes in grapevine. This includes a stronger and more prolonged oxidative burst at concentrations high enough to inhibit pathogen proliferation. To begin exploring translational applications of our findings, we also demonstrate that purified X. fastidiosa LPS elicitor can prime grapevine defenses, thereby conferring host tolerance to subsequent challenge with X. fastidiosa.

Plant cells are capable of perceiving chitin fragments (chitooligosaccharides) to trigger various defense responses. We identified a LysM receptor-like protein (AtLysM RLK1) that is required for the perception of chitooligosaccharides in Arabidopsis. Mutation of this gene blocked the induction of almost all chitooligosaccharide-responsive genes (CRGs) and led to more susceptibility to fungal pathogens, but not to a bacterial pathogen. In addition, exogenously applied chitooligosaccharides enhanced resistance against both fungal and bacterial pathogens in the wild-type plants, but not in the mutant. Together, our data strongly suggest AtLysM RLK1 is the chitin receptor or a key part of the receptor complex and chitin is a PAMP (pathogen-associated molecular pattern) in fungi recognized by the receptor leading to the induction of plant innate immunity against fungal pathogens. Since LysM RLKs were also recently shown to be critical for the perception of the rhizobial lipo-chitin Nod signals, our data suggest that LysM RLKs not just recognize friendly symbiotic rhizobia (via their lipo-chitin Nod signals), but also hostile fungal pathogens (via their cell wall chitin). These data suggest a possible evolutionary relationship between the perception mechanisms of Nod signals and chitin by plants.
