Project description:To get molecular insight into age- and compartment-specific changes in telomere maintenance and associated properties in human mammary gland, we analyzed distinct subsets of normal human mammary epithelial cells. The cells were isolated by fluorescent activated cell sorting (FACS) directly from mammary tissue obtained from normal women undergoing reduction mammoplasties with concomitant removal of hematopoietic and endothelial cells by depletion of CD45pos and CD31pos cells. The three epithelial cell populations then isolated were: (i) CD49fhiEPCAMneg/low cells, (ii) CD49fposEPCAMpos cells and (iii) CD49fnegEPCAMpos cells. The CD49fhiEPCAM-/low cells are selectively enriched in mammary stem cells with functional mammary gland regenerating activity in suitably transplanted immunodeficient mice, bipotent progenitors that form colonies of adherent myoepithelial and luminal cells in vitro, myoepithelial-restricted progenitors that form colonies of exclusively adherent myoepithelial cells in vitro, and mature myoepithelial cells that are not clonogenic (collectively referred to as basal cells, BCs). The CD49fposEPCAMpos cells are selectively enriched in luminal progenitors (referred to as luminal progenitors, LPs); and the CD49fnegEPCAMpos cells are selectively enriched in mature luminal cells (referred to as luminal cells, LCs). Differences in gene expression in general and telomere associated genes in particular were elucidated by analyzing mammary epithelial subpopulations. Total RNA was isolated from 24 samples obtained from FACS purification of mammary epithelial subpopulations from 9 reduction mammoplasty breast tissues. Project description:Age and physiological status like menopause are key factors in mammary development and are associated with breast cancer risk. Less clear is what factors influence breast cancer intrinsic subtypes that are associated with prognosis. Here, we investigated the age of the host in a mammary chimera model. The mammary glands of wildtype BALB/c mice were cleared of endogenous epithelium at 3 weeks of age and subsequently transplanted during puberty (5 weeks) or upon maturation (10 weeks) with syngeneic Trp53null mammary fragments. The Trp53null mammary epithelium undergoes a high rate of neoplastic transformation, such that carcinomas arise over the course of a year. Tumors arose more rapidly (p=0.003, log-rank test) when transplanted to adult hosts (AH, n=89) compared to tumors arising following transplantation into juvenile hosts (JH, n=55). However, JH tumors grew several times faster than AH tumors, were more perfused as indicated by 2-fold larger blood vessels (p=0.015), and exhibited a 2-fold higher mitotic index (p=0.009). Transplantation into juvenile hosts (JH) gave rise to 80% ER positive tumors compared to 60% in adult hosts. However, tumor cells were significantly more ER immunoreactive (p=0.0001) in JH tumors compared to those in AH tumors and were more highly positive for insulin like growth factor receptor phosphorylation (p=0.026). Expression profiles of JH (n=15) and AH (n=12) tumors identified 243 differentially expressed genes. The main biological processes enriched in JH tumors were organ morphology, proliferation, and lipid metabolism. An expression profile of juvenile versus adult (JVA) tumors revealed a luminal transcriptional program, characterized by expression of Foxa1, Igf2, Ccnd1 and Fgfr2. The human homologs of JVA genes (JVAh) were used to create two centroids (AHh or JHh) to classify human breast cancers from archival microarray datasets in two-dimensional space. Luminal human breast cancers were more like JH tumors compared to basal-like cancers, whereas luminal A tumors were more like JH tumors than luminal B tumors. Consistent with this, JVAh genes segregated human luminal A from luminal B breast cancers via hierarchical clustering. The distinct biology of tumors arising from tissue undergoing morphogenesis in the context of puberty suggests that young age at neoplastic transformation promotes a luminal tumor phenotype and supports the contention that the tumor intrinsic subtype is influenced by physiological status at the time of putative initiation.

Project description:One of the major hurdles for the early detection of cancer is our poor understanding of tumour initiating events. Historically, cancer research has focused on histological and molecular characterisation of established tumours which has led to the identification of hundreds of putative driver mutations. It is currently unclear how these genetic aberrations impact the cell state of nascent tumour cells and their microenvironment. BRCA1 driven triple negative breast cancer (TNBC) for example has been shown to arise from luminal progenitor cells yet little is known about how BRCA1 loss-of-function (LOF) and concomitant mutations affect the luminal progenitor cell state. This repository contains ATAC-sequencing dataset of luminal progenitors isolated from 6-months old Brca1/p53 and wild-type mice. This data was used to show that the perturbation of Brca1/p53in luminal progenitors induces an aberrant alveolar differentiation pre-malignancy. Unlike alveolar differentiation occurring during gestation, this process is cell autonomous and characterised by the dysregulation of transcription factors driving alveologenesis. The ATAC-sequencing data supports a model where transcriptional and epigenetic changes driven by Brca1/p53 inadvertently promote a differentiation program hardwired in luminal progenitors, highlighting the deterministic role of the cell of origin and offering a potential explanation for the tissue specificity of BRCA1 tumours.

Project description:SAGA and ATAC are two related transcriptional coactivator complexes, sharing the same histone acetyltransferase (HAT) subunit. The HAT activities of SAGA and ATAC are required for metazoan development but the precise role of the two complexes in RNA polymerase II transcription in mammals is less understood. To determine whether SAGA and ATAC have redundant or specific functions dependent on their HAT activities, we compared the effects of HAT inactivation in each complex with that of inactivation of either SAGA or ATAC core subunits in mouse embryonic stem cells (ESCs). We show that core subunits of SAGA or ATAC subunits are required for complex assembly, mouse ESC growth and self-renewal. Additionally, ATAC, but not SAGA subunits are required for ESC viability by regulating the transcription of translation-related genes. Surprisingly, depletion of specific or shared HAT module subunits caused a global decrease in histone H3K9 acetylation, but did not result in significant phenotypic or transcriptional defects. Thus, our results indicate that SAGA and ATAC are differentially required for viability and self-renewal of mouse ESCs by regulating transcription through different pathways, in a HAT-independent manner.
