MicroRNA profiles in KC (Kupffer cells) after partial hepatoctomy

ABSTRACT: The 70% resection promotes migration of Ly6C+CD11b+ monocytes/macrophages to the remnant liver accompanied by a reduction in its CD206+ macrophage content. mRNA and microRNA expression profiles were studied by NanoString technology and verified by a reverse transcription-based qPCR assay. Corresponding microtranscriptomic profiles cannot be unambiguously defined as pro- or anti-inflammatory. Their typical features include elevated expression of leukocyte chemoattractant factors, and many of the differentially expressed sequences are related to the control of cell growth and metabolic processes in the liver.

Macrophage proliferation within the liver reaches maximum on day 3 after the surgery. Transcriptomic profiles of macrophages isolated from the regenerating livers reveal chemokine signaling signatures in combination with indications of the involvement in specific metabolic pathways rather than inflammatory effects or morphogenesis.

Project description:Lung interstitium macrophages (IMs) are non-alveolar resident tissue macrophages which contribute to the lung homeostasis. These cells were reported to be heterogeneous by our group and other teams, which contains two main distinct subpopulations: CD206+ IMs and CD206- IMs. However, the exact origin of IMs and the transcriptional programs that regulate IM differentiation remains unclear. In recent report, we analyzed the refilled IMs in the course of time after induced IM depletion with single-cell RNA sequencing (10X Genomics Chromium) and bulk RNA sequencing. The lung IMs and monocytes from either Lyz2-Cre Mafflox/flox mice (cMAF-KO), Lyz2-Cre Mafbflox/flox (MAFb-KO), Lyz2-Cre Mafflox/flox Mafbflox/flox (dKO) or control litermate mice without floxp locus (Control) were analyzed and compared using single-cell RNA sequencing. All the main substs, i.e. Ly6C+ classical monocytes, Ly6C- patrolling monocytes, CD206+ IMs, CD206- IMs were found in control sample, while IMs are absent in both MAFb-KO and dKO sample accompied by a new intermediate population independent of Mafb expression. CMAF-KO sample show less impact in cell population composition with a lower freqency of CD206+ IM population.

Project description:As a rule, mammalian organs contain a mixed population of macrophages from different sources of origin. It is assumed that the self-sustaining resident macrophages and the monocyte-derived transient macrophages differ by their functional properties and their roles in repair processes . The analysis of the functional features of different populations of macrophages is of significant practical importance, since macrophages are considered as therapeutic targets in various pathological conditions and as prospective basis for cell therapy products. The aim of this study was to compare microRNA profiles of the two alternative macrophage species exemplified, respectively, by Kupffer cells of the liver and monocytes of the blood.

Expression in liver tissue of wildtype (wt) or hepatocyte specific Klf6 knockout (DeltaKLF6) after partial hepatectomy

Project description:KrÃ¼ppel-like factor 6 (KLF6) is a transcription factor and tumor suppressor. Loss or reduction of KLF6 is linked with progression of experimental and human hepatocellular carcinoma. Despite its important contributions to liver homeostasis and growth, there are no data characterizing the involvement of KLF6 to hepatic regeneration. Microarray data from wildtype and DeltaKlf6 mice were used to identify regulating mechanisms and potential mediators within liver regeneration Wildtype and hepatocyte specific Klf6 knockout mice (DeltaKlf6) were employed for 70% partial liver resection/hepatectomy (PHx) in order to analyse liver regeneration. Twelve hours after partial hepatectomy animals were scrificed and remnant liver tissue was used for further experiemnts. For the overall study we used 6 animals per group, and included RNA from liver tissue of 3 wildtype and 3 DeltaKlf6 animals for the microarray analysis. Wildtype animals were used as controls.

Project description:To query the relationship between miRNA profiles and deficits in liver mass after PH and the miRNAs linked to liver regeneration, we compared the miRNA expression profile after 2/3 PH, a standard procedure that leads to robust DNA synthesis, with that after 1/3 PH, a procedure that causes minimal replication. The patterns and dynamics of miRNA profiles after PH were featured, providing a rich resource of miRNAs underlying mechanisms of liver regeneration. We generated miRNA profiles from normal and remnant livers at 6, 12, 24, and 36 hours after 1/3 or 2/3PH using microarrays. Two or three rats were used at each time.

Project description:Using fluorescence activated cell sorting, we isolated CD45+, CSF1R-GFP+, F4/80+, Ly6G- mouse lung monocytes and macrophages at 7 days after pneumonectomy procedure. We then used microfluidic single cell RNA-sequencing to transcriptional profile unique myeloid subsets. Using the pneumonectomy dataset, we identified 6 cell groups and 4 gene groups that marked several regenerative macrophage subsets including CCR2+, Ly6C+ monocytes and CD206+, Chil3+ M2-like macrophages.

Mice were treated with a MAIT antagonist or its control vehicle for 2 days after the last CCl4 injection. Ly6C high and Ly6C low macrophages were sorted by ARIA III.
