Project description:Previous studies from our lab have characterized a group of splicing factors called SR proteins that function to identify intron/exon borders in regulating metabolic homeostasis in the Drosophila fat body. Decreasing the function of one SR protein, 9G8, causes an increase in triglyceride storage; however, the full complement of 9G8 target genes that regulate metabolism is unknown. To address this question, we performed RNA sequencing on Drosophila fat bodies with 9G8 levels reduced by RNAi.

Effect of transgenic RNAi on wandering third instar larval fat body gene expression in Drosophila melanogaster

Project description:We compared four transcription factor knockdowns using transgenic RNAi expressed in the larval fat body. FOXO, Tfb1, p53, and Stat92E-dependent gene expression in the Drosophila fat body was quantified on control and high-sugar diets in order to generate expression profiles via RNA-seq. These expression data were used to build a gene regulatory network to predict novel roles for these and other genes during caloric overload.

Project description:Chronic high sugar feeding induces obesity, hyperglycemia, and insulin resistance in flies and mammals. These phenotypes are controlled by the fat body, a liver- and adipose- like tissue in Drosophila flies. To gain insight into the mechanisms underlying the connection between diet and insulin sensitivity, we used Illumina RNA-seq to profile gene expression in fat bodies isolated from chronically high sugar fed, wandering (post-prandial) third instar wild type larvae w(L3). These data were compared to control-fed wild-type wL3 fat bodies as well as those expressing transgenic interfering RNA (i) targeting CG18362 (Mio/dChREBP) in the fat body on both diets.

Project description:Purpose: The goal of this RNA-Seq is to analyze the transcriptomes from wild type and TFAM RNAi fat body samples at 96 hr AEL (after egg laying) and to identify the differentially expressed genes. Methods: Drosophila larval fat bodies were isolated from wild type and TFAM RNAi (r4-gal4) animals at 96 hrs AEL (after egg laying) of development. mRAN profiles were generated by Next Generation Sequencing Facilities. Results: RNA Seq differential expression analysis identified a significant change in 1758 transcripts, with 1213 transcripts showing reduced expression levels and 545 transcripts with elevated expression levels.
