Skull and vertebral bone marrow are myeloid reservoirs for meninges and CNS parenchyma

ABSTRACT: EAE is a chronic demyelinating condition characterized by central nervous system immune infiltration. Spinal cord injury results in damage to the spinal cord, partially due to leukocyte infiltration and inflammatory contributions. Here we performed single-cell RNA-sequencing of spinal cord tissue to examine how CNS-infiltrating cells from different niches may play distinct roles in different disease pathogenesis.

Project description:Summary: Spinal cord injury (SCI) is a damage to the spinal cord induced by trauma or desease resulting in a loss of mobility or feeling. SCI is characterized by a primary mechanical injury followed by a secondary injury in which several molecular events are altered in the spinal cord often resulting in loss of neuronal function. Hypothesis: Spinal cord injury (SCI) induces a cascade of molecular events including the activation of genes associated with transcription factors, inflammation, oxidative stress, ionic imbalance, apoptosis and neuroregeneration which suggests the existance of endogenous reparative attempts. However, not all mechanisms following SCI are well known. Specific Aim: The goal of this project is to analyze the molecular events following spinal cord injury 1 cm above, below, and at the site of injury (T9), aiming at finding potential new targets to improve recovery and therapy. Analysis of the host transcriptome from demyelinating spinal cord of murine coronavirus infected mice

Project description:We use transcriptome analysis to study the spinal cord transcriptome during MHV-induced demyelinating disease and find important biological pathways for demyelinating pathology. We find evidence of a Th1 cytokine response, ongoing antigen presentation and lymphocyte proliferation, lipid metabolism changes, and eicosanoid inflammation. In addition, we report several genes important for osteoclast function have augmented expression in the CNS during demyelination, suggesting a parallel between the osteoclast and microglial functions in maintaining homeostasis and the fidelity of specialized extracellular matrices in their respective compartments. RNA-seq of mock-infected and MHV-infected spinal cord tissue at 33 days post-infection, the peak of demyelination.

miR-219 Cooperates with miR-338 in Myelination and Promotes Myelin Repair in the CNS

Project description:microRNAs (miRNAs) have been implicated in oligodendrogenesis and demyelinating diseases; however, identification of individual miRNAs responsible has remained elusive. Through targeted mutagenesis, we find that miR-219 is required for proper oligodendrocyte differentiation and myelination in vivo. Deletion of miR-338 together with miR-219 further exacerbates hypomyelination phenotypes. Temporally specific ablation reveals a critical role for miR-219 in oligodendrocyte remyelination after demyelination, while overexpression of miR-219 promotes precocious oligodendrocyte maturation and myelin regeneration. Accordingly, administration of miR-219 mimics to lysolecithin-induced demyelinating lesions in the murine spinal cord and brain enhances myelin restoration. Through integrating analyses of transcriptome profiling and biotin-affinity miRNA pull-down, we identify a set of stage-specific targets of miR-219, including Etv5 and Lingo1, as oligodendrocyte differentiation inhibitors. Inhibiting Etv5 and Lingo1 leads to a partial rescue of differentiation defects observed in miR-219-mutant OLs in vitro. Together, our findings identify context-specific miRNA-regulated checkpoints that control CNS myelinogenesis and myelin repair.

Sustained infiltration of neutrophils into the CNS results in increased demyelination in a viral-induced model of multiple sclerosis

Project description:Intracranial inoculation of the neuroadapted JHM strain of mouse hepatitis virus (JHMV) into susceptible strains of mice results in acute encephalomyelitis and chronic immune-mediated demyelination similar to the human demyelinating disease multiple sclerosis (MS). JHMV infection of transgenic mice in which expression of the neutrophil chemoattractant chemokine CXCL1 is under the control of a tetracycline-inducible promoter active within GFAP-positive cells results in sustained neutrophil infiltration in the central nervous system (CNS) that correlates with an increase in spinal cord demyelination. We used single cell RNA sequencing (scRNAseq) and flow cytometry to characterize molecular and cellular changes within the CNS associated with increased demyelination in transgenic mice compared to control animals. These approaches revealed the presence of activated neutrophils as determined by increased expression of mRNA transcripts associated with increased neutrophil effector functions, such as CD63, MMP9, S100a8, S100a9, and ASPRV1, as well as by altered neutrophil morphology and protein expression. Collectively, these findings reveal insight into changes in the profile of neutrophils associated with increased white matter damage in mice persistently infected with a neurotropic coronavirus.

The effect of upper-respiratory infection on transcriptomic changes in the CNS

Project description:Purpose: The goal of this study was to determine the effect of an upper-respiratory infection on changes in RNA transcription occuring in the cerebellum and spinal cord. post infection. Methods: Gender matched eight week old C57BL/6 mice were inoculated saline or with Influenza A (Puerto Rico/8/34; PR8, 1.0 HAU) by intranasal route and transcriptomic changes in the cerebellum and spinal cord tissues were evaluated by RNA-seq (100bp paired end reads) at days 0 (non-infected), 4 and 8 Results: After trimming and excluding multi-mappeing reads an average of 92.07% (cerebellum) and 91.71% (spinal cord) of genes were uniquely mapped to a gene. The average number of single end reads per sample was 36.42 and 37 million for the spinal cord and cerebellum respectively. Infection caused significant changes to the transcriptome of each tissue, which was most prominent at day 8 post infection. Conclusion: This study represents the first to use RNA-seq tecnology to evaluate the effect of peripheral influenza a infection on changes in gene expression of the cerebellum and spinal cord.
