Global transcriptomics of murine splenic B cells after treatment with BAFF 3mer and 60mer in presence of a control antibody or mBaffR-mFc

ABSTRACT: Mouse splenic total B cells (6 million cells/sample) were pre-treated with the control Ab (1000ng/ml, IgG1κ, BioXCell, #BE0083) or mBaffR-mFc (1000ng/ml, a fusion protein consists of murine BR3 ecto-domain fused to murine IgG1 Fc fragment obtained from Biogen, US) for 30 min, and then treated with 100 ng/ml of recombinant human BAFF 3mer (AdipoGen, #AG-40B-0016), 60mer (AdipoGen, #AG-40B-0112) or left untreated for the next 4 hours. Each treatment was in triplicates. Total RNA was extracted from the samples using QIAGEN RNeasy Mini Kit and were sent to Novogene Corporation (Wilmingtom, DE, USA) to determine the gene expression levels.

Gene expression profiling of response to Everolimus in a three-dimensional (3D) model of oral mucosa

Project description:We performed bulk RNA sequencing analysis of a 3D organoleptic model of human oral mucosa treated with everolimus. The 3D model (tissue) consisted of human normal keratinocytes grown on top of human fibroblasts and was histologically similar to buccal mucosa. The tissue was purchased from MatTek Corporation (Ashland, MA) and cultured in serum free media. The 3D model was maintained for different times in the tissue culture (40 hours and 60 hours from the time received) and treated with different concentration of Everolimus (no drug (0 ng/ml), 32 ng/mL, 64 ng/mL).

Cerebrospinal fluid ecto-GPR37 in Parkinson’s disease

Project description:Improved diagnostic and prognostic biomarkers which reflect pathological progression would benefit the management of Parkinson’s disease. GPR37, also known as parkin associated endothelin-like (Pael) receptor, is an orphan G protein-coupled receptor, which has a defective parkin-mediated ubiquitination in autosomal recessive Parkinson’s disease. By means of immunoblot and mRNA determinations we detected increased GPR37 levels in postmortem substantia nigra from Parkinson’s disease subjects. We revealed the presence of the N-terminal domain of GPR37 (ecto-GPR37) in human and mouse cerebrospinal fluid (CSF). We engineered a Nanoluciferase-based ELISA to evaluate ecto-GPR37 in CSF and demonstrated the presence of ecto-GPR37 in CSF from wild-type, but not from GPR37-/-, mice. Subsequently, the relative abundance of ecto-GPR37 in CSF in healthy controls (n=48) and Parkinson’s disease (n=45) patients was assessed. Interestingly, we found that ecto-GPR37 was significantly increased (P = 0.0002) in the CSF from Parkinson’s disease patients. Importantly, mass spectrometric analysis of immunoprecipitation of GPR37 in CSF supported the analytical validity of our ELISA to measure ecto-GPR37 in human CSF. Overall, these results open exciting perspectives and encourage further systemically studies to confirm the clinical validity and utility of ecto-GPR37 as a potential Parkinson’s disease diagnostic/prognostic biomarker
