Project description:Transcriptional profiles of a chlorhexidine tolerant Salmonella Typhimurium were compared to its, chlorhexidine sensitive, isogenic progenitor isolate. RNA was extracted from mid-log phase cells from both isolates, without chlorhexidine exposure and following exposure to 1 µg/ ml of chlorhexidine for 30 minutes. Transcriptional profiles of the tolerant isolate were compared to the sensitive isolate, with and without chlorhexidine exposure.

Transcriptomic analysis of chlorhexidine shock in Acinetobacter baumannii

Project description:In this study the transcriptomic changes occurring in A. baumannii ATCC 17978 in response to shock treament using the biocide chlorhexdine were determined. A. baumannii ATCC 17978 cells were grown at 37°C with shaking (200rpm) in 35mL cultures in MH broth to OD600=0.75, at which time they were split into 15mL cultures. One 15mL sample was treated with 4µg/mL chlorhexidine (0.5x MIC), whereas the other was not treated and used as a reference. Cultures were allowed to grow for a further 30 mins (to an average final OD600= ~1.35 for untreated cells and 1.25 for chlorhexidine treated samples), when cells were harvested by centrifugation and immediately suspended in Trizol reagent (Invitrogen). Total RNA was extracted using the PureLinkTM Micro-to-Midi Total RNA Purification kit (Invitrogen), incorporating an on-column DNAseI (Invitrogen) digestion.

Chlorhexidine promotes psl in Pseudomonas aeruginosa that enhances cell aggregation with preserved pathogenicity demonstrates an adaptation against antiseptic

Project description:Because Pseudomonas aeruginosa is a common pathogen that frequently contacts with Chlorhexidine digluconate (a regular antiseptic), then adaptations against Chlorhexidine were tested. In com-parison with the parent strain, the Chlorhexidine-adapted strain formed smaller colonies with the downregulation of several metabolic pathways (proteomic analysis) and increased resistance against colistin (an antibiotic for the current antibiotic-resistant bacteria), partly through the modification of L-Ara4N in the lipopolysaccharide at the outer membrane (proteomic analysis). Chlorhexidine-adapted strain formed dense liquid-solid interface biofilm with cell aggregation partly due to the Chlorhexidine-induced overexpression of psl (exopolysaccharide-encoded gene) through LadS/GacSA pathway (c-di-GMP-independence) in 12 h biofilms and maintained the ag-gregation with SiaD-mediated c-di-GMP dependence in 24 h biofilms as evaluated by polymerase chain reaction (PCR). The addition of Ca2+ in the Chlorhexidine-adapted strain facilitated several Psl-associated genes, indicating an impact of Ca2+ in Psl production. The sessile Chlorhexi-dine-treated bacteria demonstrated a lower expression of IL-6 and IL-8 on fibroblasts and mac-rophages than the parent strain, indicating less inflammatory reactions from the Chlorhexidine use. However, the Chlorhexidine-treated bacteria induces similar severity in 14 days of mouse wounds as indicated by wound score and bacterial burdens. In conclusion, Chlorhexidine induced psl over-expression and colistin cross-resistance that might be clinically important.

Aberrant expression of the DCT gene is implicated in the hyperpigmentation of the visceral peritoneum of the chicken

Project description:Hyperpigmentation of the visceral peritoneum (HVP) has recently garnered significant attention within the poultry industry due to its potential implications for the health of afflicted animals and its adverse effects on the appearance of commercial chicken carcasses. Unfortunately, the heritable characteristics of HVP are not yet fully understood. Motivated by poultry industry demand, we deployed multi-omics comparing the clinical HVP and normal samples profiling the potential regulating molecular landscape in cell and genome resolution, specifically, aiming to decipher the precise molecular genomic marker. Specifically, we identified the signature genes for the HVP phenotype and found that the signature gene of melanogenesis strongly enriched in the HVP group. At the melanocyte level, overexpression of the DCT gene can promote the proliferation of melanocytes, with exogenous Tyr addition. On the contrary, significantly lower melanin content in the supernatant of cells was detected after siRNA interference relative to untreated cells. Through single-cell sequencing, the hyperexpression of melanogenesis-associated genes in melanocytes was echoed. In summary, molecular features of the HVP phenotype were systematically revealed, which may partially explain the strong heterogeneities of the peritoneum phenotype. The hyperpigmentation of the peritoneum is attributed to an excess deposition of melanin, where the DCT hyperexpression can be regarded as a molecular marker related to the deposition of peritoneal melanin. Future directions should deploy gene editing technology to alert the gene expression of DCT to change the phenotype of pigmentation in the visceral peritoneum.
