MiRNA profiling in mouse peritoneal macrophages from macrophage-specific NFATc3 transgenic mice and their control littermates

ABSTRACT: We generated NFATc3 macrophage-specific transgenic mice (NFATc3MTG) using the piggyback Transposon. Peritoneal macrophages were isolated from NFATc3MTG mice and their control littermates (wild-type), and the miRNA expression profile was analyzed with deep sequencing.

Project description:To explore the roles of macrophage NFATc3 and ASK1 in metabolic dysfunction-associated steatohepatitis, we generated macrophage-specific Nfatc3 knockout mice (Nfatc3-MKO), macrophage-specific Ask1 knockout mice (Ask1-MKO),macrophage-specific double knockout (MDKO) mice, macrophage-specific Nfatc3 knockin mice (Nfatc3-MKI), and macrophage-specific Ask1 knockin mice (Ask1-MKI). Lyz2-Cre mice were used as control. Eight-week-old male Lyz2-Cre, Nfatc3-MKO, Ask1-MKO, and MDKO mice were treated with methionine- and choline-deficient (MCD) diet for 4 weeks. Subsequently, mouse liver tissues were isolated and subjected to RNA sequencing analysis. In addition, mouse bone marrow-derived macrophages (BMDMs) were isolated from Lyz2-Cre, Nfatc3-MKI, and Ask1-MKI mice and were also subjected to RNA sequencing analysis.

Project description:In this study, we aim to investigate the role of vascular smooth muscle cell (VSMC) NFATc3 in the development of AAD and establish NFATc3 as a novel target to treat AAD. We used VSMC-specific NFATc3 knockout mice in AAD model of BAPN treatment. The molecular mechanisms underlying NFATc3 function were investigated using RNA-seq analysis on aortas with BAPN treatment.

Gene expression in IRF3-/- peritoneal macrophage infected with VSV

Project description:IRF3 is one of the most critical transcription factor in down stream of pattern recognition receptors (such as toll-like receptor and RIG-I-like receptor) signalling pathway. IRF3 is known to induce the expression of type I IFN gene upon virus infection. To furter examine the role of IRF3 in virus-induced gene expression, we preformed microarray analysis in IRF3-/- peritoneal macrophages infected with VSV, and found that IRF3 suppresses the expression of Il12b gene. Peritoneal macrophages from WT of IRF3-/- B6 mice were infected with VSV(1 M.O.I. ) for 6 hous, and then subjected to microarray analysis.
