Project description:To assess the role of LSD1 in mouse small intestinal epithelium, we isolated small intestinal crypts and villus from wild type (WT) (Villin-Cre -; Lsd1f/f) and intestinal-epithelial-specific knock-out (cKO) (Villin-Cre+; Lsd1f/f) mice. This experiment uses a new Cre strain with 100% deletion efficiency. RNA was directly isolated from intestinal crypt and villus, and this was used for RNAseq. Gene expression analysis of cKO derived crypt and villus provides a spatially restricted outlook on the maturation status of the intestinal epithelium in the villi and the absence of Paneth cells in the crypt.

Project description:The intestinal epithelium is the most rapidly self-renewing tissue in adult mammals. We have recently demonstrated the presence of approximately six cycling Lgr5+ stem cells at the bottoms of small intestinal crypts1. We have now established long-term culture conditions under which single crypts undergo multiple crypt fission events, whilst simultanously generating villus-like epithelial domains in which all differentiated cell types are present. Single sorted Lgr5+ stem cells can also initiate these crypt-villus organoids. Tracing experiments indicate that the Lgr5+ stem cell hierarchy is maintained in organoids. We conclude that intestinal crypt-villus units are self-organizing structures, which can be built from a single stem cell in the absence of a non-epithelial cellular niche. Keywords: expression profiling Freshly isolated small intestinal crypts from two mice were divided into two parts. RNA was directly isolated from one part (RNeasy Mini Kit, Qiagen), the other part was cultured for one week according to the conditions described in the associated paper, followed by RNA isolation. We prepared labeled cRNA following the manufacturer’s instruction (Agilent Technologies). Differentially labelled cRNA from small intestinal crypts and organoids were hybridised separately for the two mice on a 4X44k Agilent Whole Mouse Genome dual colour Microarrays (G4122F) in two dye swap experiments, resulting in four individual arrays.

Project description:Literary fiction has been credited with considerable power to improve attitudes toward outgroups. It was even argued that it has been an important factor behind the global decline of violence against various minorities in the last centuries. Could it also help to reduce the human-inflicted suffering of animals? To test this, we studied the attitude toward animal welfare of n = 921 (experimental group) people of both sexes who read a short fragment of an unpublished novel with a motif of the physical abuse of an animal. The control group (n = 912) read a fragment of a similar length but not related to animals. After reading the text all subjects filled out an on-line questionnaire with seven items (camouflaged among many others items) measuring attitudes toward animal welfare. The questionnaire included also demographical questions, such as whether the subject keeps pets. We found that in comparison with the control group, the experimental group was significantly more concerned about animal welfare. This result indicates that literary fiction can influence attitudes toward other species. It is also worth noting that our study is characterized by a high level of ecological validity, i.e. a relatively high extent to which its results can be generalized (or extended) to real-world settings. Due to its specific design, which involved the cooperation of a bestselling author and his publisher, the study approximated the typical conditions in which people read fiction in a remarkably accurate way. Finally, our research has potential practical implications for promoting animal welfare.

Project description:Differentiation and specialisation of epithelial cells in the small intestine is regulated in two ways. First, there is differentiation along the crypt-villus axis of the intestinal stem cells into absorptive enterocytes, Paneth, goblet, tuft, enteroendocrine or M-cells, which is mainly regulated by WNT. Second, there is specialization along the cephalocaudal axis with different absorptive and digestive functions in duodenum, jejunum and ileum that is controlled by several transcription factors such as GATA4. However, so far it is unknown whether location-specific functional properties are intrinsically programmed within stem cells or if continuous signalling from mesenchymal cells is necessary to maintain the location-specific identity of the small intestine. By using the pure epithelial organoid technique, we show that region-specific gene expression profiles are conserved throughout long-term cultures of both mouse and human intestinal stem cells and correlated with differential Gata4 expression. Furthermore, the human organoid culture system demonstrates that Gata4-regulated gene expression is only allowed in absence of WNT signalling. These data show that location-specific function is intrinsically programmed in the adult stem cells of the small intestine and that their differentiation fate is independent of location-specific extracellular signals. In light of the potential future clinical application of small intestine-derived organoids, our data imply that it is important to generate GATA4-positive and GATA4-negative cultures to regenerate all essential functions of the small intestine. Project description:A comprehensive understanding of current Chinese public attitudes toward farm animal welfare and the relevant influencing factors is essential for improving farm animal welfare and promoting further development of animal husbandry. The attitudes of 3,726 respondents in China were investigated using paper and online questionnaires. Three components (affective, cognitive, and behavioral) of attitudes toward farm animal welfare were assessed using 18 items designed based on the literature review. Influential factors of attitudes toward farm animal welfare were explored via tobit regression. The results revealed that the Chinese public not only considers farm animals to be emotional and sentient but are also sympathetic toward farm animals that suffer inhumane treatment. Although they have limited knowledge about farm animal welfare, the public believes improving farm animal welfare is beneficial, especially for food safety and human health. The Chinese public prefers regulation policies to incentive policies for improving farm animal welfare. The main factors influencing attitudes toward farm animal welfare included gender, age, education, monthly household income, area of residence, farm animal raising experience, and attention to farm animal welfare events. The effect of these influencing factors on attitudes varied. These findings provide a basis for improving Chinese public attitudes toward farm animal welfare. The implications of formulating and implementing effective policies to improve the Chinese public attitudes toward farm animal welfare were discussed.
