Project description:The blockage of GABA-A ionotropic channels by means of gabazine is a widespread model of plasticity where the increased synaptic activity triggered by Gabazine leads to the up-regulation of a plethora of activity-dependent genes. Here, we sought to characterize the overall transcriptional response of GABA-A blocking of rat hippocampal organotypic cultures. In each of the three biological replica, half of the organotypic hippocampal cultures derived from two rat littermates were treated with gabazine and compared to the untreated ones in order to determine the set of differentially expressed genes.

Generation of esophageal organoids and organotypic raft cultures from human pluripotent stem cells.

Project description:The human and murine esophagus have some substantial differences that limit the utility of mouse as a model to study human esophagus development and disease. Due to these limitations several recent reports describe the development of methods to generate human esophageal tissues via the directed differentiation of pluripotent stem cells. Methods for differentiation are based on knowledge of years of studying embryonic development of the esophagus in vertebrate animal models. Esophageal tissues derived from human pluripotent stem cells have been used to study both development and diseases affecting the esophagus. Here, we provide a detailed protocol for the directed differentiation of human pluripotent stem cells into human esophageal organoids and organotypic raft cultures, that are highly similar, morphologically and transcriptionally, to the human esophagus epithelium. We discuss limitations of the current esophageal models and the importance of engineering more complex tissue models with muscle and enteric nerves. Moving forward, these models might be utilized for the development of personalized treatments, as well as other therapeutic solutions.

Effect of Molidustat on organotypic cultures of hamster cerebellum

Project description:Airborne RNA viruses from the Paramyxoviridae family include major human pathogens, in particular measles virus (MeV) and Nipah virus (NiV), which is on the blueprint list of WHO due to its high lethality rate and critical risk of emergence. Although an effective vaccine provides protection against MeV, this is not the case for NiV. Besides, there is no cure to treat MeV- or NiV-infected patients despite the risk of developing acute respiratory infections and lethal encephalitis. To identify host-targeting antivirals against these viruses, a library of metabolic modulators has been screened against MeV. Results showed that Molidustat, Daprodustat and Roxadustat, three pharmacological inducers of hypoxia-inducible factors (HIFs), have inhibitory effects on MeV infection both in vitro and ex vivo in organotypic cultures of hamster cerebellum. In addition, we showed that Molidustat inhibits NiV infection in this ex vivo model. Taken together, our results provide evidence that pharmacological induction of the hypoxia response pathway has inhibitory effect on MeV and NiV, suggesting that HIF-inducing drugs are of potential interest for treating these infections.
