Tissue-resident memory T cells (TRM) Interactions within the Tissue Ecosystem Network [scRNA-seq]

ABSTRACT: We aim to understand here, the influence of TCR-Vγ5+ γδT cells in the recruitment of TRM population. Along these lines we will analyse the populations of TRM recruited to the Skin after the challenge process (induction of allergic contact dermatitis), in the presence of TCR-Vγ5+ or TCR-Vγ5− (replacement population).

Project description:Numerous lymphocytes seed the body surfaces of metazoans promoting tissue protection and integrity. These tissue-resident populations differ substantially from circulating lymphocytes, as they adopt a unique phenotype and do not recirculate. They span the innate-adaptive continuum, ranging from innate lymphoid cells (ILCs) to unconventional T cells (NKT, MAIT, γδT cells and IELs) and tissue-resident memory (TRM) T cells, and while differ in the particulars of their biology, all contribute to barrier immunity, tissue homeostasis, and immune regulation. Although they share the same microenvironment, little is known about how these tissue-resident lymphocyte populations interact with each other and with the non-lymphoid cellular environment. As such, there is an emerging view of tissues as varied ‘‘ ecosystems ’’, whereby protection against inflammatory challenges is orchestrated through the interplay between tissue-resident and infiltrating-lymphocyte subtypes. In mice, barrier tissues are populated during early life by innate-like γδT lymphocytes. These cells provide tissue protection, including limiting conventional T cell-driven inflammation. Therefore, elucidating the contributions of tissue-resident T cells, particularly γδT cells, to the basic pathophysiology of tissue function and protection against infection, inflammation, and cancer is a priority. Tissue-Resident Memory T Cells in Human Kidney Transplants have alloreactive potential

Project description:The extent to which tissue-resident memory T (TRM) cells in transplanted organs possess alloreactivity is uncertain. This study investigates the alloreactive potential of TRM cells in kidney explants from four patients who experienced severe acute rejection leading to graft loss. Alloreactive T-cell receptors (TCRs) clones were identified in pre-transplant blood samples through mixed lymphocyte reactions, followed by single-cell RNA and TCR sequencing of the proliferated recipient T cells. Subsequently, these TCR clones were traced in the TRM cells of kidney explants, which were also subjected to single-cell RNA and TCR sequencing. The proportion of TRM cells expressing an alloreactive TCR in the four kidney explants varied from 0% to 9%. Notably, these alloreactive TCRs were predominantly found among CD4+ and CD8+ TRM cells with an effector phenotype. Intriguingly, alloreactive clones were present not only in recipient-derived TRM cells but also in donor-derived TRM cells, constituting up to 4% of the donor population, suggesting the presence of self-reactive TRM cells. Overall, our study demonstrates that T cells with alloreactive potential present in the peripheral blood prior to transplantation can infiltrate the kidney transplant and adopt a TRM phenotype.

An unconventional integrin b3 and CD103 heterodimer improves the anti-tumor immunity of tissue-resident T cells

Project description:Current cancer immunotherapies promote recovery of CD103+ tissue-resident memory T cells (Trm) population of the tumor-infiltrating T lymphocytes (TILs). However, not all treated patients exhibit improved anti-tumor immunity and survival, likely due to the immunophenotypical diversity among the CD103+ Trm TILs. Utilising multifaceted proteomics approaches and patients’ clinical analyses, we discovered an unusual subset of CD8+ Trm TILs expressing non-canonical integrin β3 early during T cells activation. The integrin β3 surprisingly heterodimerises with CD103 on T cells, leading to unconventional granulysin-mediated cytotoxicity, elevated alternative bioenergy usage and efficient T cell migration, with minimal overall exhaustion. Importantly, early-stage non-small cell lung carcinoma (NSCLC) patients with enriched presence of integrin β3+CD103+ Trm TILs exhibited better clinical prognosis, with improved T cell immunophenotype, hence confirming the beneficial role of this unusual subset of Trm TILs. These unconventional anti-tumor T cell features provide new avenues and future opportunities for designing better translational immunotherapy strategies.

Methods: CD8+THy1.1+ T cells were sorted from DLN, skin lung and liver of MAV mice and bulk RNA-sequenced Results: Heterogenouse CD8 memory populations with both circulatating and resident phenotypes were identified. Conclusions: Tumor-specific CD8 T cells form TRM responses in tumor-draining LNs.

Single cell CITE-seq and VDJ sequencing of CD8+ central memory blood T cells (TCM) and their in-vivo matured tissue-resident memory-like T cells (TRM)

Project description:The aim of the experiment was to understand the clonal relationship between CD8+ central memory blood T cells (TCM) and the in-vivo matured TCM tissue-resident memory-like T cells (TRM) on day 14. TCM were FACS sorted from human PBMCs (n=5). Half of the cells were processed on day 1 and half of the cells of each donor were matured into TRM with anti-CD3, IL-15 and TGF beta for 14 days. Both day 1 TCM and matured day 14 TRM-like cells were further processed using the 5´10X genomics workflow.
