Transcriptome analysis of rice (Oryza sativa) infected with blast fungus (Magnaporthe oryzae)

ABSTRACT: Rice blast is one of the most serious diseases and is caused by Magnaporthe grisea. SHZ-2, an indica cultivar with broad spectrum resistance to multiple races of the blast pathogen, was crossed to TXZ-13, a blast susceptible but high-quality variety, to produce one BC3 line, BC10 line, which showed strong to moderate blast resistance over eight cropping seasons in the field. Expression profiling of a lesion mimic genotype, splNF4050-8, that confers blast resistance in rice

Project description:Lesion mimic mutants in rice are widely known as spotted leaf (spl) mutants, of which several genotypes exhibit enhanced resistance to different races of Magnaporthe grisea. Besides naturally occurring spotted leaf mutants, tissue culture-induced reverse genetic repositories also act as sources of lesion mimic mutants in rice. We systematically evaluated a large collection of Tos17 mutant panel lines, developed and maintained at the National Institute of Agrobiological Sciences, Tsukuba, Japan, for their reaction to three different races of M. grisea and identified a lesion mimic mutant, NF4050-8, that showed lesions similar to naturally occurring spl5 mutant and enhanced resistance to all three blast races tested. Microarray analysis of ~44,000 rice genes in NF4050-8 with Nipponbare as control during the progressive lesion appearance stage revealed significant up-regulation of numerous defense/pathogenesis-related genes as well as several WRKY domain-containing genes and down-regulation of haem peroxidase gene. Subsequent real-time PCR analysis of WRKY45 and PR1b genes in NF4050-8 and spl5 suggested possible constitutive activation of a defense signaling pathway downstream of Salicylic Acid (SA) but independent of NH1 in these mutant lines of rice.

Rice gene global expression analysis upon inoculation with different Magnaporthe isolates

Project description:Magnaporthe oryzae is the causative agent of the rice blast, the most relevant rice disease worldwide. To date expression analysis on rice infected with Magnaporthe oryzae have been carried out only with the strains FR13 (leaf) and Guy 11 (root). However different strains of Magnaporthe are present in the environment leading to different rice responses at molecular level. To gain more insight on the unknown molecular mechanisms activated by different Magnaporthe strains during rice defense, a global expression analysis was performed by using the GeneChipÂ® Rice Genome Array. To identify rice genes differentially regulated upon infection by Magnaporthe isolates, inoculation with different strains were performed and samples were collected 24 hours post infection. RNA were obtained from leaf samples after inoculation of rice 2 week-old plantlets with the following strains: rice isolates Magnaporthe oryzae FR13 and CL367, non-adapted strain BR32, isolated from wheat, and Magnaporthe grisea BR29 isolated from crabgrass. Treated and control (mock) rice leaves (cv. Nipponbare) were collected 24 hours post inoculation. Three biological replicates for each interaction type and the corresponding mock were extracted and analysed independently with the GeneChipÂ® Rice Genome Array.

Project description:We report that the homeobox trasncription factor called HOX7 controls hyphae-associated genes, autophagy and cell cycle related genes necessary for appressorium development in the rice blast fungus Magnaporthe oryzae. We also report that the ste transcription factor MST12 regulates gene functions involved in septin reorientation.

Germination and appressorium formation in Magnaporthe grisea

Project description:This series examines differential gene expression during germination and appressorium formation by the rice blast fungus Magnaporthe grisea. Conidia were germinated on either the hydrophobic (inductive) or hydrophilic (non-inductive) side of GelBond. RNA was extracted from conidia and following incubation for 7 and 12 hours. RNA from two biological replications of each treatment was pooled in equal amounts and labeled with both cy3 and cy5 dyes using the Agilent Technologies Low Input Linear Amplification Kit. Hybridizations were performed on Agilent Technologies whole genome oligo rice blast arrays (product G4137A) using manufacturer prescribed protocols and reagents in an interlaced loop design in which each treatment was paired with every other. This series contains a total of 10 hybridizations; each treatment was used in 4 hybridizations (2 with cy3 and 2 cy5). Spot fluorescence was normalized using Lowess within and between microarray slides in Bioconductor and gene expression profiles analyzed in GeneSpring. Keywords: other
