Effect of senktide administration on the embryo collected from superovulated cows

ABSTRACT: To clarify the effect of intravenous senktide administration on the quality and developmental stage of collected embryo from superovulated cows, mRNA expression profiles of the embryo were investigated. Hierarchical cluster analysis with the expression levels of all genes was divided these cows into three clusters. First cluster was composed of control cows, second cluster contained senktide (30 nmol/min, 2h) treated cows and third cluster consisted of senktide (300 nmol/min, 2h) treated cows.

Association between progesterone concentration in early diestrus and endometrial gene expression in grazing dairy cows

Project description:The objective of this study was to evaluate the association between progesterone concentration (P4) at day 4 of the estrous cycle (beginning of diestrus) and endometrial global gene expression at day 9 (middle diestrus) in lactating grazing dairy cows. Blood samples were obtained on days 0, 4 and 9 for P4 measurement by chemiluminescence. Cows were asignated to one of the following groups (n=3/group): cows with low physiological P4 at day 4 (PLP4), cows in anestrous (ANE), cows with high physiological P4 at day 4 (HPP4) and superovulated cows (SO). Endometrial biopsy samples were obtained on day 9 for RNA sequencing. Quality control and determination of differentially expressed genes (DEG; FDR<0.05) were determined using the edgeR package for R. The number of DEG between groups were: HPP4 vs LPP4: 453; HPP4 vs ANE: 623; SO vs LPP4: 603; SO vs ANE: 1102; LPP4 vs ANE: 261 and SO vs HPP4: 0. Functional analysis using the DAVID database revealed that endometrial genes upregulated by low P4 are enriched in the immune system and inflammatory response. Conversely, genes upregulated by high P4 are enriched inmodulation of uterine relaxation-contraction, GnRH signaling pathway, focal adhesion and EGF-like related terms. In conclusion, our results support the concept of changes in P4 at the beginning of the luteal phase could be associated with the endometrial expression of critical genes involved in the preparation of the uterine environment for embryo implantation.

RNA-seq study on the placentomes collected from the cows with or without retained placenta

Project description:We conducted an RNA-sequencing on the placentomes that were collected from 4 and 3 cows with and without retained placenta, respectively. Placentomes were collected from the cows within 30 minutes after parturition and samples were taken from the feto-maternal junction sites of the placentomes. These samples were used for RNA-sequencing and the results showed 240 genes that were differentially expressed between the two groups. These genes could be candidate genes that are involved in the development of placental retention in cattle.

Project description:This project is meant to document the effect of the negative energetic balance on the embryo quality and as it happens, to identify ways to improve the fertility of dairy bovines. Measurement of the β-Hydroxybutyric acid (BHB) has been taken between 45 and 60 days post-partum on twelve Holstein cows. Cows have been ranked according to the sanguine measurement as high or low in BHB in order to get 6 cows per group. After having synchronized the heat with hormones and an ovarian stimulation, cows have been inseminated and the embryos were selected as so to be transferred in at least two heifers. This project aim at isolating the differences created on the transcriptomic expression in morulas and blastocysts grown in cows with an energetic inbalance.

Whole genome bisulfite sequencing of bovine morula derived from metabolically divergent dairy cows

Project description:Lactating cows experience transient metabolic stresses during postpartum which results in abnormal concentrations of non-esterified fatty acids and beta-hydroxybutyrate that can affect embryo-maternal communication and ultimately reproductive success. We hypothesize that metabolic challenges in lactating cows influence DNA methylation changes of the embryos prior to implantation affecting genes involved in embryo developmental competency. Therefore, we compared whole genome bisulfite sequencing (wgbs) of morulas derived from in vitro produced 2-4 cell embryos and transferred in the oviduct of metabolically profiled lactating cows until day 7. Similar stage of in vitro production (IVP) embryos were transferred to the oviduct of nulliparous heifers as a metabolically unchallenged control. Bisulfite-Seq DNA libraries were generated from groups of five morulas using an EZ DNA methylation-direct kit and Pico-Methyl seq library preparation kit (Zymo Research), and parallel sequenced using the illumine HiSeq2500 system at the institute of Genome Biology, Leibniz-Institute for Farm Animal Biology (FBN), Dummerstorf, Germany.
