Project description:Analysis of MOLT-4 cells at various time points up to 6 hours following treatment with mouse anti-CD47 antibody (MABL) and goat anti-mouse IgG (GAM) as the crosslinker of MABL. MABL induces apoptosis in CD47-positive MOLT-4 cells. Cell death signals via CD47 ligation were analyzed by using Affymetrix Human Genome U133A microarray. MOLT-4 cell line was obtained from the American Type Culture Collection (ATCC) and grown in RPMI1640 medium (SIGMA) with 10% FBS(JBS). It was maintained at 37 degree C and 5% CO2. Cells were washed with PBS(-) followed by treatment with 10 ug/mL MABL (anti-CD47 antibody) for 0, 0.5, 1, 3 and 6 h. Total RNA was isolated and converted to cDNA using a Superscript Double Stranded cDNA synthesis kit (Invitrogen). Fragmented cDNA was hybridized to Affymetrix Human Genome U133A array and gene expression signals were analyzed. Probes showing >2 or <0.5 fold-change for both replicates compared with the control (geometric mean of 0h replicate signals) were selected at any time.

Project description:Chromosomal instability (CIN), a state in which cells undergo mitotic aberrations that generate chromosome copy number variations, generates aneuploidy and is thought to drive cancer evolution. Although associated with poor prognosis and reduced immune response, CIN generates aneuploidy-induced stresses that could be exploited for immunotherapies. Macrophages, particularly, have been understudied in the CIN context. Here, through MPS1 inhibition-induced CIN in poorly immunogenic B16F10 mouse melanoma, we find that CIN- afflicted cancer cells skew macrophages towards an anti-cancer phenotype while also pushing them away from a pro-cancer one. We confirm these findings via RNA-sequencing, protein expression, and short-term tumor studies. These results further translate to in vivo efficacy in suppressing tumor growth: Mice can survive challenges of CIN-afflicted tumors. Long-term survival, however, is dependent on CD47 expression and IgG opsonization. Mice bearing CIN- afflicted tumors with wild-type CD47 levels see prolonged survival compared to their chromosomally stable counterparts, but all succumb. Mice bearing CIN-afflicted CD47 knockout tumors, however, show 28% long-term survival. When CD47 knockout was further paired with IgG opsonization, survival rate increased to 97%. Successful rejection and clearance of CIN- afflicted tumors induced de novo anti-cancer IgG antibodies that were multi-epitope and functionally promoted macrophage-mediated phagocytosis. These de novo IgG antibodies could also suppress in vitro tumoroid and in vivo tumor growth in a CD47 knockout context. These results highlight an unexpected therapeutic benefit from CIN when paired with maximal macrophage anti-cancer activity: an anti-cancer vaccination-like antibody response that can lead to durable cures and further potentiate cell-mediated acquired immunity.

Project description:Engagement of the ICOS receptor represents a key event in a process that culminates in Bcl6 expression and acquisition of the TFH and TFR phenotype. To better understand the essentials of ICOS-mediated signaling pathway, we profiled the changes in gene expression elicited after co-ligation of ICOS and CD3 compared with CD3 ligation alone. Naïve CD4+ T-cells were purified from single cell suspensions of B6 spleen and stimulated with anti-CD3 and anti-CD28 for 2d followed by resting overnight before 20 min of incubation with anti-CD3 and/or anti-ICOS and cross-linking with goat anti-hamster Ab for 8h. RNA was prepared, amplified, labeled and hybridized to Mouse Gene 1.0 ST Array (Affymetrix), in quadruplicates for anti-CD3 ligation and duplicates for anti-CD3/ anti-ICOS co-ligation.

Project description:Variations of hepatopancreatic transcript expression related to induced molt cycle were identified in male Cherax quadricarinatus using a cDNA microarray estimated to contain 2180 unique sequences. Molt induction was performed by X-organ sinus gland extirpation or by repeated 20-hydroxyecdysone injections. Manipulated males were sacrificed at premolt or early postmolt and a reference population at intermolt. Their isolated hepatopancreatic mRNA was hybridized onto the microarray and differentially-expressed genes were identified, sequenced, and annotated. Clusters of similarly expressed transcripts, in correlation with the four combinations of induction methods and molt stages were identified, containing both known and novel genes. Biologically interesting clusters were characterized by general shift of expression throughout premolt and early postmolt vs. intermolt, or by different premolt vs. postmolt expression. Several genes were differentially expressed in 20-hydroxyecdysone injected crayfish vs. X-organ sinus gland extirpated ones. Keywords: Physiological state analysis

Project description:DLD-1 and MOLT-4 cell lines were cultured in a Rotating cell culture system to simulate microgravity and mRNA expression profile was observed in comparison to Static controls Simulated microgravity affected the solid tumor cell line DLD-1 markedly which showed a higher percentage of dysregulated genes compared to the hematological tumor cell line, MOLT-4. Microgravity affects the cell cycle of DLD-1 cells and disturbs expression of cell cycle regulatory gene networks. Multiple microRNA host genes were dysregulated and significantly, mir-22, tumor suppressor microRNA, is highly upregulated in DLD-1.

Project description:DNA damage induction by either radio- or chemo-therapy has been the most widely used approach in cancer therapy, exploiting the genomic instability of cancer cells. A promising strategy takes advantage of tumour specific abnormalities in DNA damage response. Inhibition of the ATR/Chk1 pathway has been shown to be synthetically lethal in cells with high levels of oncogene-induced replication stress and in p53- or ATM- deficient cells. In the presented study, we aimed to elucidate molecular mechanisms underlying radiosensitization of MOLT-4 cells by VE-821, a higly potent and specific inhibitor of ATR. We combined multiple approaches: cell biology techniques to reveal the inhibitor-induced phenotypes, and quantitative proteomics, phosphoproteomics, and metabolomics to comprehensively describe drug-induced changes in irradiated cells.
