Project description:The olive fruit fly Bactrocera oleae is the major pest of olive production. The fly is dependent upon the symbiotic bacterium Candidatus Erwinia dacicola for the survival of the larvae in unripe olives, and in the adult stage they enhance fecundity. A major site of symbiont colonization is the esophageal bulb, yet the molecular interactions within this tissue remain poorly understood. To investigate host gene expression in this symbiont-bearing organ, we performed a transcriptomic analysis of B. oleae esophageal bulbs. This dataset provides a foundational resource for elucidating the functional biology of this organ and offers insights into insect–microbe symbiosis.

Project description:Α reduction of pyrethroid efficacy has been recently recorded in the olive fruit fly Bactrocera oleae, the most destructive insect pest of olives worldwide. We analyzed the transcriptomic differences between two highly pyrethroid resistant populations versus a relatively susceptible field population and two laboratory strains to gain more insight into the molecular mechanism of resistance. A large number of genes was found to be significantly differentially transcribed across the pairwise comparisons between resistant and susceptible insect populations. Interestingly, gene set analysis revealed that genes of the ‘electron carrier activity’ GO group were enriched in one specific pairwise transcriptomic comparison. As P450 monooxygenase enzymes are typically associated with this Molecular Function GO-group, this might reflect a P450-mediated resistance mechanism. These results suggest that transcriptional induction of the CYP6 P450s might be an important mechanism of pyrethroid resistance in B. oleae and pave the way for the development of synergists and molecular diagnostics for insecticide resistance management.

Project description:Fertility depends on the coordination of complex interactions between male and female reproductive proteins inside the female reproductive tract (FRT). These interactions mediate a suite of changes in female behavior, morphology, and physiology after mating, yet little is known about how the molecular environment of the FRT may differ among species and coordinate species-specific female post-mating responses. We used semi-quantitative proteomics to compare the FRT protein composition between virgin and mated females in Drosophila melanogaster. These results are compared to those from quantitative TMT proteomic analyses of the mating-induced changes in D. simulans and D. mauritiana, and after conspecific and heterospecific inseminations. Our study highlights the value of using quantitative proteomics approaches to study the molecular composition of the FRT environment, and how its divergence may inform mechanistic studies of post-mating pre-zygotic reproductive isolation between species.

Project description:In Drosophila melanogaster, mating radically transforms female physiology and behavior. Post-mating responses include an increase in the oviposition rate, a reduction in female receptivity, and an activation of the immune system . The fitness consequences of mating are similarly dramatic – females must mate once in order to produce fertile eggs, but additional matings have a clear negative effect. Previously, microarrays have been used to examine gene expression of females differing in their reproductive status with the aim of identifying genes influenced by mating. However, since only virgin and single mated females were compared, transcriptional changes associated with reproduction (under natural selection) and the effects of male-induced harm (under sexually antagonistic selection) cannot be disentangled. We partitioned these fundamentally different effects by instead examining the expression profiles of virgin, single mated and double mated females. We found substantial effects relating to reproduction and further effects that are only attributable to a second mating. Immune response genes dominate this male-induced harm effect indicating that the cost of mating may be due partly to this system's activation. We propose that both sexually antagonistic and natural selection have been important in the evolution of the innate immunity genes, thereby contributing to the sexual dimorphismand rapid evolution at these loci. Project description:Seminal fluid contains some of the fastest evolving proteins currently known. These seminal fluid proteins (Sfps) play crucial roles in reproduction, such as supporting sperm function, and – particularly in insects – modifying female physiology and behaviour. Identification of Sfps in small animals is challenging, and often relies on samples taken from the female reproductive tract after mating. A key pitfall of this method is that it might miss Sfps that are of low abundance due to dilution in the female-derived sample or rapid processing in females. Here we present a new and complimentary method, which provides added sensitivity to Sfp identification. We applied label-free quantitative proteomics to Drosophila melanogaster male reproductive tissue – where Sfps are unprocessed, and highly abundant – and quantified Sfps before and immediately after mating, to infer those transferred during copulation. We also analysed female reproductive tracts immediately before and after copulation to confirm the presence and abundance of known and candidate Sfps, where possible. Results were cross-referenced with transcriptomic and sequence databases to improve confidence in Sfp detection. Our data was consistent with 124 previously reported Sfps. We found 8 high-confidence novel candidate Sfps, which were both depleted in mated versus unmated males and identified within the reproductive tract of mated but not virgin females. We also identified 31 more candidates that are likely Sfps based on their abundance, known expression and predicted characteristics, and revealed that four proteins previously identified as Sfps are at best minor contributors to the ejaculate. The estimated copy numbers for our candidate Sfps were lower than for previously identified Sfps, supporting the idea that our technique provides a deeper analysis of the Sfp proteome than previous studies. Our results demonstrate a novel, high-sensitivity approach to the analysis of seminal fluid proteomes, whose application will further our understanding of reproductive biology.

In addition, we used CO2-narcosis, which induces oviposition without mating, as an experimental control for the mating process. Mating produced significant changes in the expression of vision, chemo-reception, metabolic, and immune-related genes. Differential expression of these genes maps clearly onto known behavioural and physiological changes that occur during the transition from being a virgin queen to a newly-mated queen. A subset of these changes in gene expression were also detected in CO2-treated queens, as predicted from previous physiological studies. In addition, we compared our results to previous studies that used microarray techniques across a range of experimental time-points. Changes in expression of immune- and vision-related genes were common to all studies, supporting an involvement of these groups of genes in the mating process. However, these comparisons also indicate the need to understand the temporal dynamics of gene expression across the entire mating and reproductive process.

Project description:The strictly monophagous olive fruit fly, Bactrocera oleae, represents the major pest of olive orchards worldwide. It has the unique ability to hydrolyze olive proteins as well as to overcome olive defenses, especially the high levels of phenolic compounds present in the green olive mesocarp. In this study, we aimed to identify specific genes potentially implicated in overcoming green olive defense and the utilization of the flesh, by examining larval responses to green olives on the transcript level. Focusing on the up-regulated gene set, we identified two putative serine proteases and one putative UDP-glycosyltransferase possibly associated with these traits. Serine proteases could be involved in the digestion of dietary proteins but also could represent a mechanism to overcome the effect of trypsin inhibitors induced by the olive fruit upon attack. UDP-glycosyltransferase may be implicated in the sequestration and/ or direct detoxification of phenolic compounds highly present in green olives.
