Gene expression levels of evolved thermotolerant yeast and its parent under fermentation condition

ABSTRACT: Purpose: To explore the differentially expressed genes and pathways in the thermotolerant mutant compare to wildtype. To figure out the contribution of these genes and pathways to high protein expression ability in the mutant. Method: the samples were collected from the 60h fermentation culture. The fermentation medium was BMGY/BMMY with 1%(v/v) methanol as inducer. the cultivation condition was set under 28℃ and 37℃. Results: after data analysis, we found genes related to oxidative stress response were important.

Project description:Samples for RNA-seq were collected from cell cultures in YPD medium under 30 °C or 37 °C at the logarithmic phase. The gene expression levels of thermotlerant yeast and the parent under these two temperatures were analyzed. We found the most upregualted genes of the parent under high temperature versus normal temperature were involved in autophay. the peroxisome pathway was significantly upregulated in G14 under high temperature versus the parent under normal temperature. The antixidant system of yeast was activated under thermal stress.

Comparative analysis of heat-induced genes between thermotolerant and thermosensitive tomato cultivars (50°C 1 h)

Project description:Gene expression in response to severe heat stress (50°C 1 h) was compared between thermotolerant and thermosensitive tomato cultivars. The thermosensitive cultivar showed severe impairment of heat stress response at this condition, while the thermotolerant cultivar were less affected. The results of this experiment were used to select expression marker genes that reflect the cellular heat shock response and correlate with thermotolerance.

Liquid C1 fermentation differential proteomic analysis of Eubacterium limosum

Project description:Some acetogenic bacteria, such as Eubacterium limosum, have the native ability to consume liquid C1 feedstocks, such as formate and methanol, as the sole substrate for growth. Due to high energy efficiency, and compatibility with existing infrastructure, this has sparked interest in the biotechnology industry. Previously, we reported limitations of this metabolism in batch fermentation. Here we undertook chemostat differential analysis to highlight key features and bottlenecks of metabolism. Our work serves as a reference dataset to advance understanding of liquid C1 metabolism in acetogens.

Project description:The fermentation culture of Clostridium beijerinckii mutant BA101 was monitored from exponential growth to stationary phase. During this period the culture underwent a shift from acidogenesis to solventogenesis. Acetone and butanol production was initiated with the onset of the solventogenic phase. Using DNA microarray changes in gene expression were examined during the transitional period. RNA samples were taken from Clostridium beijerinckii mutant BA101 fermentation culture at individual time points during the acidogenic phase and the solventogenic phase. The samples were used for microarray hybridization.
