Hoxd12 is indispensable for somatic cells reprogramming and proliferation of embryonic stem cells


ABSTRACT: Somatic cells reprogramming and by exogenous factors requires transcription factors coordinate with each other, but its precise mechanisms still poorly understood. Here, we report that knockdown of Hoxd12 by short-hair RNA interference (shRNA) specifically suppress 7F (Sall4-Jdp2-Esrrb-Kdm2b-Mkk6-Nanog-Glis1) induced reprogramming, nevertheless overexpression of Hoxd12 result in no significant difference of induced pluripotent stem cells (iPSCs) generation. Furthermore, knockout of Hoxd12 hinder embryonic stem cells (ESCs) proliferation, while does not affect differentiation and pluripotent. RNA sequencing (RNA-seq) reveal that Hoxd12 may affect ESCs proliferation by regulated biosynthesis-related genes. Altogether, these results suggest that Hoxd12 is necessary for efficient reprogramming of 7F and ESCs proliferate normally, this may be useful for understanding transcription factors regulatory mechanisms.
