Transcriptional profiling of Caco2 cells infected with Encephalitizoon intestinalis

ABSTRACT: Encephalitizoon intestinalis infection occurs in immunocompromised individuals and has relatively few treatment options due to the lack of understanding of impact of microsporidia life cycle on host molecular effects. We profiled transcriptome of a physiologically relevant intestinal epithelial cell line to understand what cellular processes are most impacted.

Project description:Microsporidia have attracted much attention because they infect a variety of species ranging from protists to mammals, including immunocompromised patients with AIDS or cancer. Aside from the study on Nosema ceranae, few works have focused on elucidating the mechanism in host response to microsporidia infection. Nosema bombycis is a pathogen of silkworm pM-CM-)brine that causes great economic losses to the silkworm industry. Detailed understanding of the host (Bombyx mori) response to infection by N. bombycis is helpful for prevention of this disease. The 23 K silkworm genome array was used to investigate host responses (i.e., Bombyx mori) occurring at 2, 4, 6 and 8 d post-infection by Nosema bombycis.We focused on elucidating the mechanism of the host response to microsporidia infection, especially for the investigation of host immune response . The third instar molted silkworm larvae were in oral infected by Nosema bombycis. In order to known the silkworm host response to Nosema bombycis infection at different time points, samples of infected larvae (i.e., the treatment set) and uninfected larvae (i.e., the control set) were collected at 2, 4, 6 and 8 dpi for RNA extraction and array hybridization. The obtained data were usd to investigate on the interplay of the genome-wide expression profile of hosts.

In this study, we made use of a large-scale oligo-DNA microarray to further investigate and identify genes with specific or correlated expression profiles. This large-scale oligo-DNA microarray for C. intestinalis should facilitate the understanding of global gene expression and gene networks during the development and ageing of a basal chordate. Nucleus-targeted effector EnP1 promotes microsporidia proliferation by obstructing H2B monoubiquitination and preventing host ferroptosis

Project description:Microsporidia, significant intracellular eukaryotic pathogens, pose a substantial threat to immunocompromised individuals. Their manipulation of host cells during infection remains poorly understood. Here, we identified EnP1, a previously reported spore wall protein, as a nucleus-targeted effector of microsporidia by a proximity biotinylation-based approach. EnP1's translocation to the host nucleus is meditated by nuclear localization signals (NLSs), where it interacts with host histone H2B upon infection. This interaction disrupts H2B monoubiquitination, subsequently impacting p53 expression. Crucially, this inhibition of p53 weakens its control over the downstream target gene SLC7A11, enhancing the host cell's resilience against ferroptosis during microsporidia infection. This favorable condition promotes the proliferation of microsporidia within the host cell. These findings shed light on the molecular mechanisms behind microsporidia's secretion of nuclear effectors and their control over host cell ferroptosis, ensuring their survival within the host.
