Differentially expressed genes (DEGs) analysis in control and hsa_circ_0004287 knockdown THP1-derived macrophages

ABSTRACT: To explore the detailed mechanism by which hsa_circ_0004287 inhibits M1 macrophage activation, we performed RNA-seq in control and hsa_circ_0004287 knockdown THP1-derived macrophages, and identified 43 upregulated and 35 downregulated genes with the criterion of |FC|≥2 and p≤0.05.

Project description:Expression data from untreated or Dll4-Fc treated THP1 cell line. We used Dll4-Fc stimulation of AML cells to study whether Notch activation has an impact on AML. We analyzed THP1 cell line in vitro treated with Dll4-Fc or vehicle control to determine genes affected by Notch activation. THP1 cell line was cultured on plate coated with 30 nM Dll4-Fc or vehicle for 48 hours prior to RNA extraction and hybridization to Human Genome U133 Plus 2.0 Affymetrix arrays.

MiR-511 dependently expressed genes in monoblastic cell line THP1

Project description:To experimentally investigate the function of hsa-miR-511-5p in AML monoblasts, we employed miRNA mimic mediated up-regulation of miR-511 in THP1 cells and subsequently analyzed a comprehensive gene expression profile. 4 samples of two independent miRNA mimic experiments were analyzed. THP1 monoblasts were transfected with 30 nM of Ambion Pre-miR miRNA Precursors (hsa-miR-511-5p AM10237 or negative Control #1 AM17110, Life Technologies).

Transcriptional profiling of THP1-derived human macrophages with different stimuli

Project description:Macrophages are key cell types of the innate immune system regulating host defense, inflammation, tissue homeostasis and cancer. Within this functional spectrum diverse and often opposing phenotypes are displayed which are dictated by environmental clues and depend on highly plastic transcriptional programs. Among these the ‘classical’ (M1) and ‘alternative’ (M2) macrophage polarization phenotypes are the best characterized. Understanding macrophage polarization in humans may reveal novel therapeutic intervention possibilities for chronic inflammation, wound healing and cancer. Systematic loss of function screening in human primary macrophages is limited due to lack of robust gene delivery methods and limited sample availability. To overcome these hurdles we developed cell-autonomous assays using the THP-1 cell line allowing genetic screens for human macrophage phenotypes. To confirm the relevance of the THP1-based system we performed microarray studies on THP1 cells and primary human cells subjected to various conditions. THP1 cells were treated with PMA (phorbol 12-myristate 13-acetate) to derive macrophages which were then cultured 1, 3 or 6 days with different stimuli.

Project description:The Nitroreductase NfsB (NTR) prodrug of an LSD1 inhibitor is inactive in wild-type THP1 cells, but the prodrug gets activated in the LSD1 inhibitor by the NTR in NTR-expressing THP1 cells (THP1-NTR+). Cell-type specific inhibition of LSD1 in THP1-NTR+ cells with a NTR prodrug is shown with the expression data of the two cell lines, THP1-wt and THP1-NTR+. Data for the LSD1 inhibitor and a negative control with both cell lines are included.
