Reveal seasonal climatic variations affect terpenes, norisoprenoids, and green leaf volatiles accumulation in four grape varieties under a double cropping system

ABSTRACT: We determined the RNA sequence of V. vinifera cv. Victoria and V. vinifera cv. Muscat Hamburg grapes to reveal the transcriptomics variations between summer and winter berries under a double cropping system. Transcriptomics analysis showed that the upregulated VviDXSs, VviPSYs, and VviCCDs expressions might contribute to accumulations of terpenes or norisoprenoids in winter berries.

Project description:A double cropping system has been commercially adopted in subtropical regions in southern China, where there is abundant sunshine and heat resources. In this viticulture system, the first growing season normally starts as a summer cropping cycle; then, the vine is pruned and forced by hydrogen cyanamide, resulting in a second crop in January of the next year. Due to climate differences between the two growing seasons,the transcriptome varies greatly between summer- and winter- grape berries. In this study, we conducted RNA-seq for two table grape varieties (Vitis vinifera cv. Victoria and V. vinifera cv. Muscat Superior) at three developmental stages (pre-veraison, post-veraison and full maturity).The transcriptome between summer- and winter- grape berries were compared.

Transcription profiling of grape berries during ripening

Project description:Grape berries undergo considerable physical and biochemical changes during the ripening process. Ripening is characterized by a number of changes, including the degradation of chlorophyll, an increase in berry deformability, a rapid increase in the level of hexoses in the berry vacuole, an increase in berry volume, the catabolism of organic acids, the development of skin colour, and the formation of compounds that influence flavour, aroma, and therefore, wine quality. The aim of this work is to identify differentially expressed genes during grape ripening by microarray and real-time PCR techniques. Using a custom array of new generation, we analysed the expression of 6000 grape genes from pre-veraison to full maturity, in Vitis vinifera cultivar Muscat of Hamburg, in two different years (2006 and 2007). Five time points per year and two biological replicates per stadium were considered. To reduced intra-plant and inter-plant biological variability, for each ripening stadium we collected around hundred berries from several bunch grapes of five plants of V. vinifera cv Muscat of Hamburg. Differential protein expression in berry skin from red grapes with varying hybrid character

Project description:Protein expression from berry skin of four different red grape biotypes was compared at a proteome-wide level by bottom-up shotgun proteomics, label free quantification and MaxQuant-assisted computational analysis. Red grapes were from a purebred Vitis vinifera (Aglianico cv.), a V. vinifera (local Sciascinoso cv.) grafted onto an American rootstock, an interspecific hybrid (V. vinifera × V. labrusca, Isabel) and an uncharacterized red grape with some hybrid lineage, as demonstrated by the presence of relatively high amounts of anthocyanidin 3,5-O-diglucosides. The aim was assessing the differences among red grape biotypes at a protein expression levels, also addressing the possible effect of the grafting on the phenotypic expression of some key metabolic enzymes in grape berries.

Project description:Plant roots secrete secondary metabolites to sense the enviroment around them. Among them, terpenes play a prominent role. Terpenes can have either fungistatic or fungicide action. However, their exact role in plant-host interactions is not fully understood. Verticillium longisporum is a soilborne pathogen causing disease in Brasicacae plants. In this project we investigated the transcriptomic changes of this species upon exposure to the β-pinene monoterpene in different time points 0hpi, 8hpi, 24hpi and 24hpi.

Expression data in individual grape berries during ripening initation

Project description:We used Affymetrix microarray analyses of thirty-two individual Vitis vinifera cv. Cabernet Sauvignon berries sampled from two clusters at fifty-percent ripening initiation. By delineating four developmental stages of ripening initiation, we demonstrate that color is a statistically significant indicator of transcriptional state during ripening initiation. We report on clustered gene expression patterns which were mined for genes annotated with signal transduction functions in order to advance regulatory network modeling of ripening initiation in grape berries. We also demonstrated that gene expression does not differ statistically significantly at a global level in berries sampled from different plants or different positions in a cluster. Effect of temperature and irradiation intensity on grapevine berry ripening in Muscat Hamburg fruiting cuttings

Project description:At the onset of ripening (veraison), Vitis vinifera L. cv Muscat Hamburg fruiting cuttings were exposed to four different environmental regimes in two growth chambers set up: 20/15ºC (day/night) and 30/25ºC (day/night) and with two different photosynthetic photon flux density (PPFD) conditions (200 and 400 μmol•m−2•s−1 PPFD) at grape bunch level during a 14 h photoperiod. Fruiting cuttings were selected to have similar grape bunch size avoiding changes in berry quality. At least 9 plants were transferred to each treatment. Global pericarp gene expression was compared among the four growth conditions using the GrapeGen GeneChip®. The transcriptomic analysis was carried out in 14 DAT berries in order to identify stable responses to differential temperature and PPFD conditions not masked by quick transient responses to the environmental change.
