RNA-sequencing of adipocyte and stromal vascular cells (SVCs) of epididymal and inguinal adipose tissue from normal chow diet (NCD)-fed mice

ABSTRACT: In mammals, white adipose tissues are largely divided into visceral epididymal adipose tissue (EAT) and subcutaneous inguinal adipose tissue (IAT) with distinct metabolic properties. To investigate molecular mechanisms underlying depot-specific metabolic roles, we report the transcriptomes of adipocytes and SVCs derived from NCD-fed mouse epididymal adipose tissue (EAT) or inguinal adipose tissues (IAT).

Project description:We report the transcriptomes of adipocytes and SVCs derived from HFD-fed obese mouse epididymal adipose tissue (EAT) or inguinal adipose tissues (IAT). By the comparisons of transcriptome of adipocyte and SVC from EAT and IAT, we find that the gene clusters of gamma-aminobutyric acid (GABA) signaling among the three-hundred fourteen pathways annotated in the Kyoto Encyclopedia of Genes and Genomes (KEGG) database was differentially regulated in SVCs from IAT compared to SVCs from EAT. Q-RT-PCR analyses confirmed the reliability and high degree of sensitivity of gene the RNA-sequencing results. Further, when the SVC from IAT with the SVC from EAT were compared, six pathways were included in the Pareto optimal set, excluding pathways below 1.0 and –log10(0.05) for the x and y-axis, respectively. Selected pathways and their differentially expressed genes were visualized as a bipartite graph using Cytoscape software to reveal that differentially regulated GABA signaling pathways could be associated with immune cell adhesions.

Project description:Committed preadipocyte fibroblasts were genetically labelled in transgenic mice by expressing GFP under the control of the locus for Zfp423, a gene controlling preadipocyte determination. These mice are herein referred to as Zfp423-GFP mice. The overall goal was to identify genes differentially expressed between adipogenic GFP+ firboblasts and non-adipogenic GFP- fibroblasts from either inguinal or epididymal fat stromal vascular cultures obtained from Zfp423-GFP mice. Adipose stromal vascular cultures were obtained from inguinal or epididymal fat of 6-8 week-old male Zfp423-GFP mice. GFP+ and GFP- fibroblasts were sorted from fat stromal vascular cultures and then returned to culture. Subconfluent cultures of GFP+ and GFP- cells were compared in triplicate for Affymetrix analysis.

Project description:Visceral white adipose tissue is closed correlated with obesity and metabolic dysfunction. Epididymal adipose tissue (eWAT) is considered as typical visceral white adipose tissue. Induction of browning of white adipose tissue improves metabolic dysfunction such as insulin resistance. In contrast to mice subcutaneous adipose tissue, visceral fat do not show significant browning under 4°C. However,under physiologically tolerable low temperature visceral adipose tissue can turn brown. We used microarrays to detail the global programme of gene expression in C57Bl/6 mice epididymal adipose tissue exposed to thermoneutral 30°C, 4°C and temperatures lower than 4°C.

Inguinal white adipose tissue expression profiles of WT mice and Slc35d3 adipose tissue knock-in mice

Project description:Slc35d3 adipose-specific knockin (SAKI) mice were generated by crossbreeding mice carrying the Slc35d3 transgene locus controlled by loxp-stop-loxp sequence (LSL-Slc35d3) with adiponectin-derived Cre mice, which is a mouse model that expresses the cre enzyme specifically in adipose tissues. Male 6-week-old Slc35d3 adipose-specific knockin mice (SAKI group) and their control mice (wild-type mice, WT group) were fed a normal standard chow diet (NCD, 10 kal% fat, D09100304, Research Diets, Inc) for 10 weeks. All mice reached the experimental endpoint at 16 weeks of age. Mice in WT group were used as controls. Upon reaching the experimental endpoint, inguinal white adipose tissue (IngWAT) from three SAKI mice and three WT mice were removed for RNA sequencing.
