Identification of the differentially expressed genes in the mouse embryonic dorsal spinal cord from E10.5 to E12.5

ABSTRACT: To identify differentially expressed genes in the developmental mouse dorsal spinal cord, we characterized the global gene expression profiling of mouse embryonic dorsal spinal cord commissural neurons at E10.5, E11.5 and E12.5. We used the Affymetrix Mouse Exon 1.0 ST Array platform to analyze the gene expression profiling. We included the gene expression data obtained from dorsal spinal cord commissural neuron at different embryonic stage. 2 Biological replicates were performed.

RNA sequence of embryonic dorsal skin and AGM (Aorta-gonad-mesonephros) tissues

Project description:Purpose: The goals of this study are to identify the different gene expresion level in dorsal skin cells or AGM region at different embryonic stages Methods: Dorsal skin cells were collected from different-staged embryo: E9.5, E10.5 E12.5 and E14.5. AGM were collected from different-staged embryo: E9.5, E10.5 and E11.5. In duplicate. RNA were extrated and analyzed using RNA sequencing. For dorsal skine tissues, we screened the genes were highly expressed at early embryonic stage E9.5, but not later stages (E10.5, E12.5, and E14.5). For embryonic AGM tissues, we selected the genes were highly expressed at early embryonic stage E9.5, but not later stages (E10.5 and E11.5). The gene list were filtered based on secreted potein candidates. These gene candidates might function as novel embryonic anti-tumor factors Results: Using an optimized data analysis workflow, we mapped the gene list for potential secretory anti-tumor factors. Conclusions: One of the top gene from the gene lists is NEPN which we hypotherze it has unique anti-tumor function in the early embryonic microenvirnemt and might be used as a novel cancer therapeutic drug

Transcription profiling of mouse E14.5 dorsal spinal cord

Project description:This experiment aims at characterizing the transcriptome of embryonic mouse dorsal spinal cord. Dorsal spinal cords dissected from litters of E14.5 wild type embryos of unknown sex were processed for RNA extraction using Trizol and RNeasy Mini kit (Qiagen) extraction procedures. Five replicates of wild type embryos were analyzed, each sample with tissue pooled from three embryos.

The precursors of hematopoietic stem cells during mouse embryogenesis

Project description:Definitive hematopoietic stem cells (HSCs) bud off from the hemogenic endothelial cells (HEC) located in the dorsal aorta of the mouse embryo. The maturation of HSCs from HEC occurs through the precursor of HSCs (pre-HSCs) between embryonic day (E) 9.5 and E11.5. To clarify the differentiation process of pre-HSCs, we performed single-cell RNA-seq analysis of cells dissociated from the dorsal aorta and its surrounding tissues and the fetal liver at E10.5 and E11.5. We identified pre-HSCs population generated from an arterial marker-expressing cluster. Thus, our data might be useful to understand the differentiation process of pre-HSCs.

RNA-Seq of developing mouse eye (E11.5 and E12.5) comparing optic fissure margins with dorsal optic cup

Project description:We sought to identify gene expression signatures confined to the small group of cells at the fissure margins that are involved in OFC. Serial cryosections perpendicular to the optic fissure were prepared from mouse embryonic eyes (n=3 at E11.5 and n=3 at E12.5). The fissure margins and a corresponding control region of dorsal optic cup were isolated using laser capture microdissection. This study design aimed to identify the signature of gene expression in the OFC margins and to identify those genes that were more highly expressed along the ventral (inferior) fissure compared to the opposing dorsal (superior region). Fissure closure is active at E11.5 and complete by E12.5

Single-cell RNA-seq of NR5A1-eGFP positive cells of the developing mouse ovary from E10.5 to P6

Project description:Purpose: We performed a time-course single-cell RNA-seq of the somatic cells of the XX mouse gonads to study the cell population heterogeneity and the genetic program during their differentiation. Methods: We collected gonads from NR5A1-eGFP transgenic embryos at six embryonic stages: E10.5, E11.5, E12.5, E13.5, E16.5 and P6. Methods: Cells were capture with the C1 autoprep system and cDNA sequenced with Illumina HiSeq 2000. Results: One cell population was detected at E10.5 and give rise to both Granulosa and steroidogenic precursor cells. A precursor cell population remains undifferentiated at P6 and are likely to be theca cell precursors. Conclusion: Our study is, to date, the most granular transcriptomic study of the developing mouse ovary and provide a more complete model of somatic cell differentiation during female sex determination.
