Metaproteomes of a cyanobacterial consortium from a haloalkaline bioreactor over a 12 day dark incubation

Project description:A photosynthetic cyanobacterial/microbial consortium was incubated in the dark for 12 days. During the course of this dark incubation, samples were taken every two days from the biomass portion and the liquid (supernatant) portion of the bioreactor. Metaproteomics analysis was conducted on these time series samples and binned and assembled metagenomes from the same samples were used as the database for protein identification.

Contact-dependent killing by Caulobacter crescentus via cell surface-associated, glycine zipper proteins

Project description:Most bacteria are in fierce competition with other species for limited nutrients. Some bacteria can kill nearby cells by secreting bacteriocins, a diverse group of proteinaceous antimicrobials. However, bacteriocins are typically freely diffusible, and so of little value to planktonic cells in aqueous environments. Here, we identify an atypical two-protein bacteriocin in the gamma-proteobacterium Caulobacter crescentus that is retained on the surface of producer cells where it mediates cell contact-dependent killing. The bacteriocin-like proteins CdzC and CdzD harbor glycine-zipper motifs, often found in amyloids, and CdzC forms large, insoluble aggregates on the surface of producer cells. These aggregates can drive contact-dependent killing of other organisms, or Caulobacter cells not producing the CdzIE immunity protein. The Cdz system uses a type I secretion system and is unrelated to previously described contact-dependent inhibition systems. However, Cdz-like systems are found in many bacteria, suggesting that this form of contact-dependent inhibition is common.

Project description:Transcriptome analysis to determine the impact of oral exposure (in a sugar meal) to the liquid supernatant (i.e. LB culture media) of Chromobacterium sp. Panama biofilm culture. The biofilm supernatant (i.e. media) was first filtered with a 0.2uM filter to remove all live bacterial cells. It was then mixed with 10% sucrose, and a control sucrose meal was mixed with filtered LB. Mosquitoes were exposed to each sugar meal for 24 hours and then midguts were dissected from 20 adult females per treatment. The entire experiment was performed 4 independent times.

Development of gene expression signatures for contact-inhibited and adipogenic-differentiated 3T3-L1 and NIH3T3 fibroblasts

Project description:Given previous works showing that, in the process of adipogenic differentiation of 3T3-L1 fibroblasts, the cells need to be cultured to confluency followed by additional incubation before initiating differentiation, we hypothesized that contact inhibition of proliferation (CIP) is requisite for making the cells prone to the differentiation. We screened upregulated genes in contact-inhibited 3T3-L1 fibroblasts, as well as NIH3T3 fibroblasts that are also sensitive to contact inhibition, by a whole genome microarray analysis. We also screened the genes that undergo rapid downregulation after the initiation of adipogenic differentiation.
