Project description:Portal hypertension (PHTN) is a severe complication of liver cirrhosis. It is associated with intrahepatic sinusoidal remodeling induced by sinusoidal resistance and angiogenesis. Collagen type IV (COL4), a major component of basement membrane (BM), forms in liver sinusoids upon chronic liver injury. However, the cellular source and transcriptional regulation of COL4, as well as how it progresses in liver diseases is unknown. Here, we examined how COL4 is produced and how it regulates sinusoidal remodeling in PHTN. RNA-seq analysis on human cirrhotic livers revealed an increase in COL4 expression, which was further confirmed via immunofluorescence (IF) staining. scRNA-seq analysis identified Liver sinusoidal endothelial cells (LSECs) as the predominant source of COL4 expression in mouse liver, which was upregulated in a TNFα-NFκB dependent manner. Epigenetic repression of enhancer-promoter interaction silences COL4 gene expression via dCas9-KRAB-mediated epigenome editing approach. LSEC-specific COL4 mutation or repression abrogated sinusoidal resistance and angiogenesis, thereby alleviated sinusoidal remodeling and PHTN. Our findings reveal LSECs as a major source of COL4 in the liver which plays a prominent role in sinusoidal remodeling and PHTN.

Gene expression of liver sinusoidal endothelial cells from an 8-week-old WT and ob/ob mouse

Project description:Obesity may promote myeloid cell accumulation in liver. Intravital liver tissue imaging revealed that the number of myeloid cells attached to sinusoidal wall significantly increased in ob/ob mice. To assess the pathophysiological roll of liver sinusoidal endothelial cells (LSEC) in obese livers, we performed microarray analysis in LSEC isolated from an 8-week-old wild-type and ob/ob mouse. Pathway analysis in microarray data shows genes related to cell adhesion, chemokine signaling and leukocyte transendothelial migration are upregulated in LSEC from ob/ob.

Identification of molecular pathways involved in oxaliplatin-associated sinusoidal dilatation

Project description:Oxaliplatin-based chemotherapy for colorectal liver metastases (CRLM) can result in vascular liver lesions such as sinusoidal dilatations. Physiopathology remains unclear and variability between patients suggests that there is individual susceptibility. A better understanding of the molecular mechanisms of oxaliplatin liver toxicity may allow to identification of biomarkers and adaptation of chemotherapy delivery. non-tumor frozen liver samples were obtained from patients operated on for CRLM after an exclusive oxaliplatin-based chemotherapy. Gene-expression profiles were first analyzed by microarray on a selected population of 20 patients; 9 patients with severe sinusoidal dilatation (SD) after a short period of chemotherapy and 10 patients without any sinusoidal dilatation after a long period of chemotherapy. These were compared with a control group of 5 patients without any chemotherapy and lesions. This dataset is part of the TransQST collection.

Transcriptomic analysis of senescent cells upon knockdown of 38 candidate SASP-specific regulators.

Project description:IMR90 ER:RAS cells were transfected with scramble siRNA or 2 deconvoluted siRNAs targeting each of 38 candidate genes or positive control siRNAs targeting the senescence regulators p16 and p53 and the SASP regulators NF-kappaB, p38alpha and CEBPbeta. The next day the cells were treated with 4OHT to induce senescence. 6 days later the cells were collected for total mRNA analysis. Our previous experiments had shown that these 38 genes regulate the secretome of senescent cells without affecting other senescence phenotypes such as the growth arrest. By performing RNA-seq we confirmed that knockdown of these genes affected the expression of multiple pro-inflammatory factors in senescence. More importantly, the secreted factors were differentially regulated across the different knockdown conditions indicating potential for specific manipulation of immune response genes.
