Single Cell sequencing of tumor infiltrating immune cells upon E2 treatment

ABSTRACT: Estrogens (E2) has been shown to affect growth, differentiation and polarization of different immune cell types in different context. To understand how E2 treatment affects immune microenvironment during cancer (melanoma) progression, we profiled tumor infiltration immune cells from placebo and E2 treated tumors isolated from female mTyrCreERT2;BrafCA/WT;Ptenf/f. We identified diverse immune cell types and their underlying transcriptomic changes that are manifest upon E2 treatment in the tumor microenvironment.

Project description:Estrogens(E2) are important steroid hormones that regulate differentiation, proliferation, and apoptosis in hormone-dependent breast cancer.In order to detect the E2-dependent transcription program associated with the observed cell cycle response, we analyzed the effect of H2ac knockdown on MCF-7 gene expression using microarray. Interestingly, we noticed that 51% of the E2-upregulated genes are down-regulated by depletion of H2ac. The data also show that H2ac regulated E2-dependent genes through E2-induction signaling pathway.

RNA-seq of estrogen (E2) induced changes of mRNA expression in early pregnant Sus scrofa endometrium

Project description:Maternal exposure to estrogens can induce long-term adverse effects in the offspring. This may be mediated through alterations in the endometrium affecting embryo-maternal communication as early as the preimplantational phase. Thus, we analyzed the effects of gestational estradiol-17β (E2) exposure on the endometrium. Two distinct low doses and a high dose (0.05, 10 and 1000 µg E2/kg body weight daily, respectively) were orally applied to sows from insemination until sampling at day 10 of pregnancy and compared to carrier-treated controls. RNA-sequencing revealed a dose-dependent increase of 14, 17 and 27 differentially expressed genes (DEG), respectively. Overall, the maternal E2 treatment perturbed gene expression of the endometrium, potentially altering the uterine histotroph.
