Effect of C2S in RAW264.7 murine macrophages

ABSTRACT: This study aimed to investigate the mechanism of C2S-induced macrophagic inflammation and its’ effect on osteogenic differentiation of precursor cells. This study used the C2S (75-150 μg/ml) extract to induce macrophagic inflammation in RAW264.7 murine macrophages. RNA sequencing was preformed for analyzing mechanism of mitochondrial function and autophagy.

Project description:Hypoxia promotes macrophage polarization triggered by IL-6.In an attempt to gain speci?c insight into the mechanism of macrophage polarization during hypoxia. We used microarrays to detail the global programme of gene expression and identified distinct classes of changed genes during this process. Four groups of RAW264.7 cells were harvested for RNA extraction and hybridization on Affymetrix microarrays (normoxia control, normoxia+IL-6,hypoxia control,hypoxia+IL-6).

High-resolution transciptome mapping and N6-methyladenosine modification mapping of mouse macrophage RAW264.7 cells

Project description:Purpose: Detect transcriptome-wide mapping of m6A modifications of mouse macrophage RAW264.7 cells by MeRIP-seq after 24 h-treatment with 100 μM 2,4,6-trichlorophenol (TCP), 100 μM 2,4,6-tribromophenol (TBP) and 100 μM 2,4,6-triiodophenol (TIP). Methods: Total RNA was extracted using TRIzol Reagent. Poly (A) RNA was purified from 50 μg total RNA using Dynabeads. Then the poly(A) RNA was fragmented into small pieces using Magnesium RNA Fragmentation Module.The fragmented RNA was incubated with m6A-specific antibody. The desired m6A-enriched RNA eluate was used to construct cDNA library in parallel with input control. Conclusion: Our study revealed the difference of gene expression at transcriptional level and m6A-methylated RNA among TCP, TBP and TIP treated RAW264.7 cells by RNA-seq and MeRIP-seq technologies.

Project description:Purpose: Next-generation sequencing (NGS) has revolutionized systems-based analysis of cellular pathways. The goals of this study are to evaluate the gene expression of M2-polarized RAW264.7 cells compared with M0 RAW264.7 cells. Methods: RAW264.7 cells were polarized toward M2 using IL-4. M0 RAW264.7 cells were maintained in culture without IL-4. Total RNA of M2 and M0 RAW264.7 cells was extracted. RAW264.7 cells RNA profiles were generated by deep sequencing for two groups (M2 versus M0 RAW264.7 cells) with three samples each. Results: There were significant differences between M2 and M0 RAW264.7 cells. Conclusions: Polarization of RAW264.7 cells from M0 to M2 induces various changes at the transcription level.
