Genome-wide expression changes in response to the treatment of anti-CD93 in mouse tumor endothelial cells

ABSTRACT: We isolated ECs from anti-CD93 antibody treated or control KPC tumor and performed RNA-seq to investigate genome-wide expression changes in response to the treatment of anti-CD93. Many of the genes affected by anti-CD93 were functionally related to cell adhesion, angiogenesis or vasculature development. Gene ontology analysis further confirmed that the transcriptomic changes by anti-CD93 were mainly related to biological pathways of angiogenesis, cell adhesion, or vasculature development.

Mitochondrial respiration controls neoangiogenesis during wound healing and tumour growth

Project description:<p>The vasculature represents a highly plastic compartment, capable of switching from a quiescent to an active proliferative state during angiogenesis. Metabolic reprogramming in endothelial cells (ECs) thereby is crucial to cover the increasing cellular energy demand under growth conditions. Here we assess the impact of mitochondrial bioenergetics on neovascularisation, by deleting cox10 gene encoding an assembly factor of cytochrome c oxidase (COX) specifically in mouse ECs, providing a model for vasculature-restricted respiratory deficiency. We show that EC-specific cox10 ablation results in deficient vascular development causing embryonic lethality. In adult mice induction of EC-specific cox10 gene deletion produces no overt phenotype. However, the angiogenic capacity of COX-deficient ECs is severely compromised under energetically demanding conditions, as revealed by significantly delayed wound-healing and impaired tumour growth. We provide genetic evidence for a requirement of mitochondrial respiration in vascular endothelial cells for neoangiogenesis during development, tissue repair and cancer. </p>

Priming a vascular-selective cytokine response permits CD8+ T-cell entry into tumors

Project description:Targeting DNA methyltransferase 1 (DNMT1) has immunomodulatory and anti-neoplastic activity, especially when paired with cancer immunotherapies. Here we explored the immunoregulatory functions of DNMT1 in the tumor vasculature. Dnmt1 deletion in endothelial cells (ECs) impairs tumor growth and metastatic seeding while priming expression of cytokine-driven cell adhesion molecules and chemokines important for CD8+ T-cell trafficking across the vasculature; consequently, the efficacy of immune checkpoint blockade (ICB) is enhanced. Tumor ECs reduced in IFNg response genes are enriched in cell cycle effectors, including Dnmt1, indicating mitotic ECs suppress anti-tumor immune responses. Targeting Dnmt1 in ECs reduces proliferation and augments adhesion and diapedesis of CD8+ T-cells, suggesting Dnmt1 programs immunologically anergic tumor vasculature. Our study is in good accord with preclinical observations that pharmacologically disrupting DNMT1 enhances the activity of ICB, but suggests an epigenetic pathway presumed to be targeted in cancer cells, is also operative in the tumor vasculature.

mRNA expression in HEK-293 /STF cells

Project description:Canonical Wnt signaling in endothelial cells (ECs) is required for vascularization of the central nervous system (CNS) and for formation and maintenance of barrier properties unique to CNS vasculature. Gpr124 is an orphan member of the adhesion G-protein-coupled receptor family that is expressed in ECs and is essential for CNS angiogenesis and barrier formation via an unknown mechanism. Using canonical Wnt signaling assays in cell culture and genetic loss- and gain-of-function experiments in mice, we show that Gpr124 functions as a co-activator of Wnt7a- and Wnt7b-stimulated canonical Wnt signaling via a Frizzled receptor and Lrp co-receptor, and that Gpr124-stimulated signaling functions in concert with Norrin/Frizzled4 signaling to control CNS vascular development. Common and distinct functions of mouse Dot1l in the regulation of endothelial transcriptome

Project description:Epigenetic mechanisms are mandatory for endothelial cells (ECs) during cardiovascular development. Dot1l-mediated gene transcription in mice is essential for development and function of lymphatic ECs (LECs). The role of Dot1l in development and function of blood ECs (BECs) is unclear. RNA-seq datasets from Dot1l-depleted or -overexpressing BECs and LECs were used to comprehensively analyze regulatory networks of gene transcription and pathways. Dot1l depletion in BECs changed the expression of genes involving cell-to-cell adhesion and immunity-related biological processes. Dot1l overexpression changed the expression of genes involved in different types of cell-to-cell adhesion and angiogenesis-related biological processes. Genes involved in specific tissue development-related biological pathways were altered in Dot1l-depleted BECs and LECs. Vascular endothelial growth factor production-related genes were specifically regulated in BECs by Dot1l. Dot1l overexpression uniquely altered ion transportation-related genes in BECs and immune response regulation-related genes in LECs. Importantly, Dot1l overexpression in BECs led to expression of many genes related to the angiogenesis and mitogen-activated protein kinase signaling pathways. The expression of these genes was unchanged in Dot1l-overexpressing LECs. The findings demonstrate the unique transcriptomic program of ECs and the differential functions of Dot1l in the regulation of gene transcription in BECs and LECs.

CD93 negatively regulates astrogenesis in response to MMRN2 through transcription repressor ZFP503 in the developing brain

Project description:Astrogenesis is repressed in the early embryonic period, and occurs in the late embryonic period. A variety of external and internal signals contribute to the sequential differentiation of neural stem cell. Here, we discover that the immune-related CD93 plays a critical role in the onset of astrogenesis in mouse. CD93 expression is detected in neural stem cell and neuron but bot in astrocyte, and declines as differentiation proceeds. CD93 knockout increases astrogenesis at the expense of neuron production during the late embryonic period. CD93 responds to the ECM protein MMRN2 to trigger the repression of astrogenesis. In mechanism, CD93 delivers the signal through a series of phosphorylation cascade to β-Catenin, then β-Catenin transfers to nucleus to activate Zfp503 transcription. The transcription repressor ZFP503 inhibits Gfap transcription by binding to the Gfap promoter with the help of Grg5. Furthermore, the abnormal differentiation in astrogenesis and neurogenesis caused by CD93 knockout results in autism-like behaviors. Taken together, our study reveals that CD93 is a negative regulator in controlling the proper time of astrogenesis, and provides a new sight in therapy for psychiatric disorders.

While it is convenient to believe that HSCs represent a homogenous population, several evidences do not support this notion. Here we identified and characterized a HSC population that expressed CD93 on their membrane. This population showed strong molecular dHSC signature, combined with activation features. Thereby, suggesting that they represent an intermediate state between quiescent and active HSCs being more prone to respond upon alterations in the hematopoietic lineage
