Identification of Aryl Hydrocarbon Receptor (AhR) as a Functional Target that Enhances Astrocytic ApoE Secretion

Project description:We discovered a chemical series that enhances ApoE secretion from human astrocytes through mechanisms independent of LXR agonism. Using chemoproteomics, photoaffinity probes, KINOMEscan analysis, and siRNA knockdowns, we identified the aryl hydrocarbon receptor (AhR), a transcription factor not previously linked to ApoE secretion, as the primary target. AhR antagonism was confirmed to increase ApoE secretion through a panel of agonists/antagonists and genetic knockdown. While chronic AhR inhibition is unsuitable for Alzheimer's treatment due to peripheral effects, these findings highlight AhR as a modulator of ApoE secretion and a pathway worth further exploration. There is two datasets deposited for this project. One dataset was generated through affinity chromatography enrichment, and the second dataset was generated through photo-affinity chromatography enrichment.

Transcriptomic changes assoicated with hepatic AHR or ARNT loss in female mice challenged with high-fat diet

Project description:The aryl hydrocarbon receptor (AHR) is an important hepatic transcription factor that appears to have a role in regulating endogenous liver metabolism. Canonical AHR-mediated regulation of transcription requires AHR binding to its DNA-binding partner, the AHR nuclear translocator (ARNT). We previously demonstrated that the AHR can regulate transcription independent of ARNT via “non-canonical” interactions with Kruppel-like factor 6, and that hepatic AHR loss can protect against high-fat diet (HFD) challenge. In this study, we examined the effects of hepatic AHR or ARNT loss in HFD-fed mice to determine whether AHR regulation of liver metabolism might partially occur independent of ARNT. The data show that similar to AHR loss, ARNT loss reduces HFD-driven hepatic lipid deposition, but does not recapitulate the diminished weight gain and adiposity observed in AHR KO animals. Utilizing transcriptomic sequencing analyses, we uncovered specific gene pathways that likely account for these phenotypic differences/similarities associated with AHR or ARNT loss. Together, these data highlight the physiological significance of ARNT-independent AHR activity, and identify discrete aspects of liver metabolism likely regulated via canonical AHR action.

Genome-wide expression profile of livers from mice fed HFD,SO-HFD or Viv chow [RNAseq]

Project description:Purpose: Aim of the study is to identify changes in hepatic gene expression induced by either a 40kcal% coconut oil rich high fat diet (HFD), a 40kcal% soybean oil plus coconut oil high fat diet (SO-HFD) or a low fat vivarium chow diet (Viv). Methods: Livers from mice that had been fed one of the above mentioned diets for 35 weeks, were used to make cDNA libraries that were then sent for deep sequencing, using the Illumina TruSeq RNA. Result: Many genes involved in metabolism, lipid binding, transport and storage and many Cyp genes are dysregulated in the two high fat diets as compared to Viv HFDs in SO-HFD mice. Comparing the two HFDs shows more metabolism and disease related genes dysregulated in SO-HFD vs HFD. Conclusion: A diet high in soybean oil may be more detrimental to metabolic health than a diet high in saturated fats. cDNA isolated from livers from mice fed HFD, SO-HFD or Viv for 35 weeks, were 50bp pair-ended sequenced in triplicate using Illumina TruSeq RNA Sample Prep v2 Kit.

Gene expression in chow or HFD-fed OGG1-/- livers

Project description:Gene expression in livers of male wild-type (WT) and OGG1-deficient (Ogg1-/-) mice fed either a chow diet or a high-fat diet (HFD) were examined. Mice were fed the diet for 10 weeks prior to tissue collection and were 22 weeks of age at the time of tissue collection. 24 Total samples were analyzed. We generated the following pairwise comparisons using GeneSifter: WT Chow vs Ogg1-/- Chow; WT HFD vs. Ogg1-/- HFD using t-test followed by Benjamini and Hochberg correction. An adjusted p-value less than 0.05 was considered to be statistically significant.

Gut Microbiota Metabolite Indole-3-Acetic Acid Maintains Intestinal Epithelial Homeostasis via the transcription factor AHR

Project description:The incidence and prevalence of inflammatory bowel disease (IBD) is gradually increasing. A high-fat diet (HFD) is known to disrupt intestinal homeostasis and aggravate IBD, yet the underlying mechanisms remain largely undefined. Here, a positive correlation between dietary fat intake and disease severity in both IBD patients and HFD-fed mice is observed. HFD induces a significant decrease in indole-3-acetic acid (IAA) and lead to intestinal barrier damage. Furthermore, IAA supplementation enhances the intestinal mucin sulfation and effectively alleviates colitis. Mechanistically, IAA upregulates key molecules involved in mucin sulfation, including Papss2 and Slc35b3, the synthesis enzyme and the transferase of 3'-phosphoadenosine-5'-phosphosulfate (PAPS), via the Aryl Hydrocarbon Receptor (AHR). More importantly, AHR can directly bind to the transcription start site of Papss2. Oral administration of Lactobacillus reuteri, which can produce IAA, contributes to protecting against colitis and promoting mucin sulfation, while the modified L. reuteri strain lacking the iaaM gene (LactobacillusΔiaaM) and the ability to produce IAA fails to exhibit such effects. Overall, IAA enhances intestinal mucin sulfation through AHR, contributing to the protection of intestinal homeostasis.

RNA Sequencing of livers from ApoE-/- mice treated with HIF1a inhibitor PX-478 or Saline

Project description:Purpose: HIF1a inhibitor PX-478 prevents hypercholesterolemia in ApoE-/- mice fed Western diet for 3 months. The goal of this study was to determine the mechanisms in the liver through which PX-478 regulates cholesterol. Methods: 4-week old ApoE-/- mice were fed a Western diet and injected intraperitoneally, bi-weekly with either PX-478 (40 mg/kg) or Saline for 3 months. The livers of 3 PX-478-treated mice and 4 Saline-treated mice were used for this RNA-seq study. Results: We identified 800+ differentially expressed genes in the PX-478-treated ApoE-/- mice livers relative to the saline-treated control livers. A subset of these genes were found to play important roles in cholesterol regulation and were further validated by qPCR analysis. Conclusions: Our study details potential hepatic cholesterol regulating mechanisms for HIF1a inhibitor PX-478. These results, coupled with our findings on the effect of PX-478 on two different mouse models of atherosclerosis, demonstrate that PX-478 is a potential anti-atherogenic and anti-hypercholesterolemic drug.
