MRNA expression profile in BMCMCs from C57BL/6J and CC027 mice stimulated with or without IgE-ligation.

ABSTRACT: Collaborative Cross (CC) mice are a reproducible yet genetically diverse genetic reference panel derived from eight founder strains. Here, we compared the gene expression profiles of mast cells (MCs) from C57BL/6J (a traditional inbred strain) and CC027 (a CC strain) mice. We generated bone-marrow-derived cultured MCs (BMCMCs) from C57BL/6J and CC027 mice. The cells were sensitized with anti-DNP IgE overnight, then were stimulated with or without DNP-HSA, followed by total RNAs extraction. RNA-seq library construction and sequencing was done by Novogene (https://en.novogene.com/).

Mapping of a QTL for airway hyperresponsiveness on mouse chromosome 12

Project description:The BcA86 strain is a unique recombinant congenic strain created from parental strains A/J and C57BL/6J. Naive mice from the BcA86 strain have a lung responsiveness phenotype resembling mice from airway hyperresponsive strain A/J. However, majority of the BcA86 genome is from the hyporesponsive strain C57BL/6J. Our goal was to identify the genomic regions that are associated with this BcA86 phenotype. Using F2 mice generated from BcA86 backcrossed to C57BL/6J, we identified a QTL for airway hyperresponsiveness on mouse chromosome 12. We validated the importance of mouse chromosome 12 in airway responsiveness using a chromosome 12 substitution strain (CSS12) which contains A/J chromosome 12 on a C57BL/6J background. The CSS12 strain also had a lung responsiveness phenotype similar to A/J. We selected genes within our QTL as candidates for airway hyperresponsiveness if they contained a deleterious coding variant (based on PROVEAN analysis) or if they were differently expressed between hyperresponsive (A/J, BcA86, CSS12) and hyporesponsive (C57BL/6J) strains.

Epigenome analysis of liver samples from C57BL/6J, CD1, and Sv129Ev mice in lifelong chronic psychosocial stress (LCPS)

Project description:Genome wide DNA methylation profiling of liver tissues from 17 months old male mice belonging to either the C57BL/6J, the CD1 or the Sv129Ev strain and exposed to LCPS. The Illumina Infinium Mouse Methylation Beadchip was used to obtain DNA methylation profiles across approximately 285,000 CpGs in liver samples. Project description:To determine if genetic background can modulate severity of an infection, we studied the host responses to influenza infections in the eight genetically highly diverse Collaborative Cross (CC) founder mice. The CC founder (C57BL/6J, 129S1/SvlmJ, CAST/EiJ, PWK/PhJ) were intranasally infected with influenza A/HK/01/68 (H3N2) with 20Î¼l virus solution (1x101 ffu) or mock infected (with PBS).
