Demonstration of increased biologically active CXCL12Î± plasma concentrations after ACKR3 antagonist treatment in humans

Project description:CXCL12 is a chemokine that binds to its cognate receptors CXCR4 and ACKR3 (CXCR7). An increase in CXCL12 concentrations has been used as a pharmacodynamic biomarker to assess ACKR3 antagonism in healthy adults. Furthermore, increased CXCL12 concentrations have been observed in various human pathologies. To date, CXCL12 concentrations have typically been quantified using antibody-based assays with overlapping or unclear specificity for the various existing CXCL12 proteoforms. Only the N-terminal full-length CXCL12 proteoform is biologically active and can engage CXCR4 and ACKR3. However, this proteoform could so far not be quantified in healthy adults. Here, we describe a new and validated fit-for-purpose immunoaffinity mass spectrometry (IA-MS) biomarker assay for specific measurement of five CXCL12Î± proteoforms in human plasma, including the biologically active CXCL12Î± proteoform. The assay was employed in a Phase 1 clinical study with the ACKR3 antagonist ACT-1004-1239 to quantify CXCL12Î± proteoforms. At baseline levels, 1.00 nM total CXCL12Î± and 0.10 nM biologically active CXCL12Î± was quantified in placebo treated adults. The concentrations of both proteoforms increased up to two-fold in healthy adults following drug administration. At all doses, 10% of CXCL12Î± was biologically active and the simultaneous increase of all proteoforms suggests that a new equilibrium has been reached 24 h following dosing. Hence, this IA-MS biomarker assay can be used to specifically measure active CXCL12 proteoform concentrations in clinical trials. Specific quantification of active chemokines can support decision making in clinical trial and thus successful drug development.

Circulating CircRNA expression profiling of Graves' disease

Project description:NGS was performed to detect human circRNAs. We found many upregulated and downregulated circRNAs in GD patients. Through high-throughput sequencing technology, 23215 upregulated and 13715 down-regulated circRNAs were detected between gd patient group and the healthy control group. Of the 218 circRNAs with significant difference, 359 were up-regulated and 309 were down-regulated.

Circulating LncRNA expression profiling of Hashimoto's Thyroiditis

Project description:NGS was performed to detect human lncRNAs. We found many upregulated and downregulated lncRNAs in HT patients. Through high-throughput sequencing technology, 69,544 upregulated and 27,742 down-regulated LncRNAs were detected between HT patient group and the healthy control group. Of the 218 LncRNAs with significant difference, 94 were up-regulated and 124 were down-regulated.

Plasma circulating MiRNAs: a new potential biomarker for prostate cancer diagnosis.

Project description:Purpose: PCa is the second most commonly diagnosed malignancy in men. PCa Diagnosis are based on biopsy sampling that is an invasive, expensive procedure and does not accurately represent multifocal disease. It is desirable to have an easily accessible, minimally invasive way to accurately determine the molecular signature of patient’s tumor that can aid in diagnosis and risk stratification. Methods:we enrolled a total of 70 patients underwent 12-core transrectal ultrasound (TRUS) biopsy of which 48% had cancer in the diagnosis. The mean age at diagnosis was 67.15 (IR 55/75), the mean PSA (ng/ml) at diagnosis was 7.8 (IR 4.1/13.4) and the mean TRUS Volume (ml) was 53.5 (IR 29/86). The cancer biopsy presented 73.1% of positive core. Among all cancer patients, 57% showed a High grade of cancer status (Grade 3). Controls are patients with Benign Prostate Hyperplasia (BPH). MicroRNA-expression profiling (NGS analysis) was performed to identify tumor-related microRNAs (miRs) and determine their association with clinicopathological characteristics. Results: The expression of miRs -4732-3p, let7a, 26b-5p, 98-5p, 30c-5p and 21-5p may identified Prostate Cancer in plasma samples. Higher expression of mir-4732-3p is associated with a high grade tumor Conclusions: miRs signature discriminate Prostate cancer in plasma better to PSA values and is associated with high grade tumor status

Plasma circulating miRNA profiling in polymicrobial sepsis by high throughput sequencing

Project description:Aim: To explore the potentially important role of circulating miRNAs in systemic inflammation during sepsis. Method: 8 to 10-week old C57BL/6 mice underwent cecal ligation and puncture (CLP) or laparotomy (Sham). Plasma RNA was isolated using Trizol LS reagent 24h post-surgery. NGS cDNA libraries were prepared using Norgen Biotek Small RNA Library Prep Kit. Library quality was validated prior to sequencing on an Illumina NextSeq 500 platform.

Project description:NGS has been applied to microRNA-enriched RNA obtained from Extra Virgin Olive Oil (EVOO), beer and plasma samples of healthy volunteers that usually consume EVOO hours after the ingestion of 40 mL of EVOO. Results: We did not detect significant amount of microRNA in the EVOO samples. Plasma samples did not contain EVOO microRNAs nor other microRNAs from plant origin.

A MicroRNA Next-Generation-Sequencing Discovery Assay (miND) For Genome-Scale and Absolute Quantitation of Circulating MicroRNA Biomarkers

Project description:The plasma levels of tissue-specific microRNAs can be used as prognostic and diagnostic biomarkers for chronic and acute diseases. Thereby, the combination of diverse miRNAs into biomarker signatures using multivariate statistics seems especially powerful in view to tissue and condition specific miRNA shedding into the plasma. Although Next-Generation Sequencing (NGS) technology enables to analyse circulating microRNAs on a genome-scale level, it suffers from potential biases (e.g. adapter ligation bias) and lacks absolute transcript quantitation. In order to develop a robust NGS discovery assay for genome-scale quantitation of circulating microRNAs we first evaluated the sensitivity, repeatability and ligation bias of four commercially available small RNA library preparation protocols. The protocol from RealSeq Biosciences was selected based on its performance and usability, and coupled with a novel panel of exogenous small RNA spike-in controls to enable absolute quantitation and ensure comparability of data across independent NGS experiments. The established MicroRNA Next-Generation-Sequencing Discovery Assay (miND) was validated for its relative accuracy, precision, analytical measurement range and sequencing bias and was considered fit-for-purpose for microRNA biomarker discovery. Summarized, all these criteria were met and thus our analytical platform is considered fit-for-purpose for microRNA biomarker discovery from plasma, serum, cerebrospinal fluid (CSF), synovial fluid (SF), or extracellular vesicles (EV) extracted from cell culture medium in the setting of any diagnostic, prognostic or patient stratification need.
